血必净对大鼠铜绿假单胞菌肺炎的抗炎作用

摘　　要：目的探讨血必净注射液对铜绿假单胞菌肺部感染的抗炎作用。方法将96只大鼠随机分成模型组、抗生素组与血必净组，每组32只。通过气管插管，气管内注入铜绿假单胞菌建立小鼠肺部感染模型，抗生素组注入头孢他啶．血必净组再加用血必净注射液。分别在0、6、24、72h时间点抽取尾静脉血液，用ELISA法检测各组血浆肿瘤坏死因子-a（TNF-a）与白细胞介素-6（IL-6）的浓度，并检测血液白细胞总数。结果三组TNF-a、IL-6浓度与白细胞数在0、6h时比较差异无统计学意义；在24、72h抗生素组与血必净组较模型组三者均降低（P〈0．01）。TNF-a、IL-6浓度血必净组较抗生素组降低（P〈0．01）。但白细胞数在24h后两组间比较差异无统计学意义；在72h血必净组较抗生素组低（P〈0．05）。1周后三组病死率分别为46．9％、12．5％、6．3％。结论血必净注射液能抑制铜绿假单胞菌肺炎的炎性反应，与抗茵药同时使用可减缓病情。Objective To investigate the effect of xuebijing injection on blood inilarnmatory mediators expression in rats with pseudomonas aeruginosa pneumonia. Methods Ninty - six rats were divided into three groups randomly： model group, antibiotic group and xuebijing group （n = 32 each）. Pseudomonas aeruginosa strain was instilled into the trachea of the rats through tracheal cannula to induce pneumonia in each group, ceftizidime was instilled into in antibiotic group, and xuebijing injection was instilled into in xuebijing group in addition. Samples of blood were harvested at 6,24,72 hours after infection. The count of white blood cell（WBC） in blood and the concentrations of TNF -a and IL-6 in serum were measured by enzyme - linked immunosorbent assay （ELISA） at different time, respectively. Results At 0 and 6 h, all index were no difference among the three group. At 24 h and 72 h, All index were significantly higher in model group than those in antibiotic group and in xuebijin group （P 〈 0. 01 ） , the TNF - a and IL - 6 level were higher in antibiotic group than those in xuebijin group （P 〈 0. 01 ）. At 24 h, the count of WBC was no difference between latter group, but higher in antibiotic group than in xuebijin group at 72 h. （P 〈 0.05 ）. After a week, the mortality of rats in model group （46. 9% ） was significantly higher than that in antibiotic group （12.5%） and in control group（6.3% ）, between the latter two, antibiotic group showed higher mortality （P 〈 0. 01 ）. Conclusions Pneumonia can be ameliorated by the xuebijing injection cooperating with antibiotic suppressing inflammatory re- spone .
