摘 要:目的:研究人卵巢黄素化颗粒细胞中抗苗勒氏管激素(AMH)对干细胞因子(SCF)表达的影响。方法:收集15例年龄<35岁因男方因素行体外受精-胚胎移植(IVF-ET)患者的卵巢颗粒细胞,经原代培养,加入不同浓度基因重组人抗苗勒氏管激素(rhAMH),分别于培养的第4日与第6日采用实时定量PCR(RT-PCR)和免疫组织化学检测空白对照组与各实验组SCFmRNA及蛋白的表达。结果:RT-PCR和免疫组织化学均证实人卵巢黄素化颗粒细胞在不同浓度rhAMH干预后,SCFmRNA及蛋白的表达显著减少(P<0.05),其中15 ng/ml rhAMH抑制作用最明显;实验组第4日与第6日颗粒细胞表达SCFmRNA及蛋白无明显差异(P>0.05)。结论:AMH可有效抑制人卵巢黄素化颗粒细胞SCF mRNA及蛋白的表达。Objective: To explore the influence of anti Mullerian hormone(AMH) on the expression of SCF in human granulosa cells cultured in vitro.Methods: Fifteen patients were recruited who received in vitro fertilization and embryo transfer(IVF-ET) at age 35 years only because of male sterility,and the granulosa cells were cultured with different concentrations of rhAMH.The expressions of SCF mRNA and protein in granulosa cells were determined by RT-PCR and immunohistochemistry on day 4 and day 6,respectively.Results: The results of both RT-PCR and immunohistochemistry confirmed that rhAMH can significantly inhibit the expression of SCFmRNA and protein(P0.05) in human granulosa cells cultured in vitro.With the increase of rhAMH to 15 mg/L,the expressions of SCF mRNA was further down-regulated.There was no difference between day 4 and day 6(P0.05).Conclusion: AMH can effectively down-regulate the expressions of SCF mRNA and protein in human granulosa cells.
