胃癌组织中VEGF-D的表达与MLVD的相关性研究

摘　　要：目的:检测胃癌患者肿瘤组织血管内皮生长因子(VEGF)-D的表达及微淋巴管密度(MLVD),分析VEGF-D与MLVD的关系。方法:收集52例胃癌术后患者标本,采用免疫组化方法测定胃癌、癌旁和正常组织MLVD及VEGF-D的表达。结合临床及病理资料,分析VEGF-D与MLVD、年龄、性别、肿瘤组织分化、淋巴结转移以及分期之间的关系。结果:VEGF-D在胃癌、癌旁与正常组织的阳性表达差异有统计学意义(P<0.01),而在胃癌与癌旁组织两者间的阳性表达差异无统计学意义。有淋巴结转移和Ⅲ、Ⅳ期患者胃癌及癌旁组织中VEGF-D阳性率高于无淋巴结转移和Ⅰ、Ⅱ期患者(P<0.05或P<0.01)。癌旁MLVD高于正常组织及胃癌组织。癌旁组织中VEGF-D阳性的MLVD高于VEGF-D阴性者(P<0.01)。而胃癌组织中VEGF-D阳性的MLVD与VEGF-D阴性的MLVD差异无统计学意义(P>0.05)。结论:胃癌癌旁组织中VEGF-D及MLVD的检测可作为评估胃癌淋巴结转移、判断胃癌生物学行为及预后的指标之一。Objective： To detect the expression of vascular endothelial growth factor-D （VEGF-D） and micro-lymphatic vessel density （MLVD） in patients with gastric cancer, and their relationship thereof. Methods： Fifty-two samples of gastric cancer patients after surgery were collected. The immunohistochemistry method was used to determine expression levels of VEGF-D and MLVD in gastric cancer, borderline tumor and normal tissues. The relationship between expression levels of VEGF-D and MLVD, patient age, gender and clinical and pathological data were analysed. Results： There was a significant difference in the positive expression rate of VEGF-D between gastric cancer, borderline tumor and normal tissues （P〈0.01）, but no significant difference between gastric cancer and borderline tumor tissues. There was a higher positive rate of VEGF-D in gastric cancer patients with lymph node metastasis and in stageⅢand Ⅳ than that in patients without lymph node metastasis and in stageⅠandⅡ（P〈0.05 and P〈0.01）. The expression level of MLVD was higher in borderline tumor than that in normal tissue. The expression level of MLVD was higher in borderline tumor of patients with positive expression of VEGF-D than that of patients with negative expression of VEGF-D. There was no significant difference in the expression of MLVD between gastric cancer patients with positive expression of VEGF-D and patients with negative VEGF-D （P〈0.05）. Conclusion： The detection of VEGF-D and MLVD in tissues of gastric borderline can be one of important markers to evaluate the lymph node metastasis, the malignant biology and prognosis of gastric cancer.
