摘 要:目的:观察补肺汤对肺纤维化大鼠肺组织中基质金属蛋白-9(MMP-9)和组织基质金属蛋白酶抑制剂-1(TIMP-1)蛋白表达及肺组织病理变化的影响。方法:雄性SD大鼠,随机分成4组:空白组、模型组、强的松组、补肺汤组。除空白组外,其余3组均采用博莱霉素(BLM)制作肺纤维化大鼠模型,每日灌胃给药,造模后7、14、28天取材。通过免疫组织化学染色方法,检测肺组织MMP-9、TIMP-1蛋白表达,采用光学显微镜观察肺组织病理变化。结果:与模型组比较,补肺汤组、强的松组各时间点肺泡炎和肺纤维化程度均较轻;与模型组比较,各时间点强的松组、补肺汤组肺组织MMP-9蛋白表达明显减低;与模型组比较,各时间点的强的松组、补肺汤组肺组织TIMP-1蛋白表达明显降低。结论:补肺汤能够抑制早期肺泡炎的发生,降低肺组织MMP-9与TIMP-1蛋白表达,纠正MMP-9/TIMP-1失衡,改善细胞外基质的异常代谢,从而抑制或延缓博莱霉素致大鼠肺纤维化的发生和发展。Objective :To observe dynamically the effects of polygonum cuspidate on the expression of tumor necrosis factor - α ( TNF - α) and platelet - derived growth factor (PDGF) in the bronchoalveolar lavage fluid (BALF) of pulmonary fibrosis rats and the pathological changes of lung tissue. Methods :96 SD male rats were divided into four groups randomly including control group, model group, prednisone group and Bufei Decoction group (n = 24). Except the normal control group, other 3 groups were all adopted bleomycin(BLM) to set up rat model with pulmonary fibrosis by intragastric administration every day, and the materials were draw 7,14,28 days after making models. The contents of MMP -9 and TIMP - 1 of lung tissue were detected with with immunohistochemical staining method and the pathological changes of lung tissue were observed with optical microscope. Results : Compared with model group, the degree of alveolitis and pulmonary fibrosis in Bufei Decoction and prednisone group was mild compared with model group, the expressions of MMP -9 and TIMP - 1 were decreased significantly in Bufei Decoction and prednisone group. Conclusion:Bufei Decoction can decrease the degree of alveolitis and pulmonary fibrosis decreased significantly in the dexamethasone group and Bufei Decoction could decrease the expressions of MMP - 9 and TIMP - 1 in lung tissue and reestablish the balance between MMP - 9 and TIMP - 1, then improve the abnormal metabolism of extracellular matrix, so that it could inhibit the formation of pulmonary fibrosis.
