摘  要:目的:探讨砒霜(三氧化二砷ATO)对MRL/lpr自发性狼疮小鼠外周血单个核细胞(PBMC)中DNA甲基转移酶1(DNMT1)和黏附分子淋巴细胞功能相关抗原-1(LFA-1)的亚基CD 11a表达的影响及其可能作用机制。方法:20周龄(发病期)MRL/lpr狼疮小鼠24只,设C57BL正常小鼠24只做参照,随机平分两组,即治疗组(ATO组)和对照组(NS组),治疗组ATO 0.4mg.kg-1.d-1隔日腹腔注射,对照组隔日腹腔注射同等量生理盐水,疗程为60天,采用RT-PCR检测两组小鼠PBMC的DNMT1及CD11a转录水平的表达情况。结果:MRL/lpr小鼠治疗组中DNMT1-mRNA的灰度比值对照组增高(P<0.05),MRL/lpr小鼠治疗组中CD11a-mRNA的灰度比值较对照组降低(P<0.05)。结论:砒霜能逆转系统性红斑狼疮(SLE)的低甲基化状态。Objective: To investigate the ATO regulatm'y mechanism on the expression of DNA Methyhransferase 1 - mRNA and CD11amRNA in lupus nephritisprone MRL/lpr mice. Methods :24 MRL/lpr mice (20 weeks old )and 24 C57 BL mice were equally divided into 2 groups randomly: control group (saline , NS group) and treatment group (arsenic trioxide 0.4 mg/kg · d, ATO group) . These drugs were given by abdominal injection for 60 days. The expression of DNMT1 and CD 1 la was determined by RT - PCR. Results : The gray scale value of DNMT1 - mRNA in ATO group was significantly higher than that in NS group( P 〈 0.05 ) in MRL/lpr mice , and the gray scale value of CD11 a - mRNA in ATO group was significantly lower than that in NS group ( P 〈 0.05 ) in MRL/lpr mice. Conclusion : The mechanism of ATO of the treatment of SLE may be related with reversed low methylation.
