十全大补汤对Lewis肺癌小鼠MMP-9基因表达的影响

摘　　要：目的:探讨十全大补汤对Lewis肺癌组织中MMP-9基因表达的影响。方法:50只小鼠采用细胞株接种的方法复制Lewis肺癌转移模型,造模成功后,将小鼠分为模型组,CTX组,十全大补汤低、中、高剂量组。采用RT-PCR方法检测十全大补汤对Lewis肺癌组织中MMP-9基因表达的影响。结果:十全大补汤中、高剂量组的肿瘤转移抑制率为43.20%、29.59%,两组中MMP-9的表达下降,与模型组比较有统计学意义(P<0.01)。结论:十全大补汤下调MMP-9基因表达是其抑制肺癌转移的分子机制之一。Objective ： To study the effect of Shiquan Dabu Decoction on MMP - 9 gene expression in Lewis lung cancer mice. Methods ： The 50 mice were randomly divided into 5 groups ： the model group, the Cyclophosphamide group, the low,middle and high dosage groups of Shiquan Dabu Decoction after the cancerometastasis mice model of Lewis lung cancer were established by inoculating tumor cell. The gene expression of MMP - 9 in tumour tissues were detected by reverse- transcription polymerase chain reaction（ RT- PCR）. Results： The tumor metastasis inhibition rate in the middle and high dosage groups of Shiquan Dabu Decoction were 43.20% and 29.59% respectively. Moreover, the contents of MMP-9 genes,compared with model group, were decreased （ P 〈0.01 ）. Conclusion： Reducing gene expression of MMP -9 was one of molecular mechanism of Shiquan Dabu Decoction inhibiting lung cancer＇ s metastasis.
