摘  要:目的建立非冻存低温条件下维持悬浮培养细胞的细胞保存技术。方法常规培养悬浮生长的人白血病细胞Jurkat,分别正常培养、室温和4℃冰箱条件下保存,间隔10天进行细胞形态观察和细胞存活率分析,然后室温和4℃冰箱保存的细胞再加入新鲜培养剂转入正常培养。结果两种非冻存低温条件下细胞存活率降低,与正常培养组有显著性统计学差异(P<0.01),并且在细胞形态上具有相应的改变,10天后存活率分别为81%和79%,没有差异(P>0.05),20天后室温条件下细胞仍然保持81%存活率,与前10天没有差异(P>0.05),并在转入正常培养条件下恢复生长繁殖。结论非冻存低温条件下可以短期维持悬浮培养细胞,该方法丰富了细胞培养技术。Objective To establish a technique of maintaining suspension cells under a non-freezing low temperatures.Methods A suspension culture cell line,Jurkat(human leukemia cells),was placed in room temperature and 4℃ environment respectively and These cells were measured on cell survival rate and shapes every 10 days before growing under normal condition.Results Cell survival rates of two non-freezing low temperature groups were decreased and had a significant difference(P0.01) compared with the control.Meanwhile,the cell shape was changed.But compared with the control,there was no statistical difference(P0.05) on survival rates(81% and 79%) after 10 days treatment.81% of cells still survived after 20-day treatment of room temperature and had no difference(p0.05) compared with 10-day treatment group,which still normally grew in standard culture condition subsequently.Conclusion Short-term culture of Suspension culture cells can be short-term cultured in non-freezing low temperatures.This method enriches the knowledge of cell culture.
