去卵巢大鼠脑内MDA水平、GSH-Px活性及NO浓度的变化及半枝莲黄酮的干预作用  被引量:2

摘  要:目的:去卵巢大鼠脑内MDA水平、GSH-Px活性及NO浓度的变化及半枝莲黄酮(Scutellaria Barbata flavonoid,SBF)对这些变化的影响。方法:3月龄SD雌性大鼠摘除卵巢10个月后,用分光光度法检测大脑皮质和海马中丙二醛(malonyldialde-hyde,MDA)、一氧化氮(nitric oxide,NO)的含量及谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)的活性,并观察SBF对这些变化的影响;结合雌激素倍美力为阳性对照药。结果:与假手术组大鼠相比,双侧卵巢摘除大鼠大脑皮质和海马MDA水平分别增加了0.58倍和0.50倍、GSH-Px活性分别降低了40.13%和57.53%,NO含量分别降低了49.25%和44.90%。然而去卵巢大鼠连续灌胃SBF 17.53,57,0 mg.kg-1 36天可不同程度地改善去卵巢大鼠上述神经异常变化,0.065 mg.kg-1倍美力也表现出类似的作用。结论:双侧卵巢摘除可以引起大鼠脑内MDA、NO水平与GSH-Px活性异常改变,而SBF可以对抗上述异常变化,提示SBF这种神经保护作用有利于神经退行性疾病的治疗。OBJECTIVE To study the changes of MDA level,GSH-Px activity NO content in ovariectomized rats and the interventional effect extrted by flavonoids from Scutellaria Barbata (SBF). METHODS 3 months SD female rats were ovariectomized for 10 months, and evaluated the effects of SBF on MDA, NO level and GSH-Px activity in cerebral cortex and hippocampus of ovariectomized rats using spectrophotography. Premarin was taken as the reference drug. RESULTS Compared with sham group, in the ovariectomized rats the level of MDA increased 0. 58 and 0. 5 folds, the activity of GSH-Px lowered 40. 13% and 57. 53%, and the content of NO decreased 49.25% and 44. 90% in cerebral cortex and hippocampus, respective ly. However, these deteriorated changes in ovarietomized rats were improved by SBF 17. 5, 35,70 mg·kg-1 administered daily for consecutive 36 days. The reference drug premarin also revealed the similar results. CONCLUSION The ovariectomization with rats can cause the abnormal changes of MDA level, GSH-Px activity and NO content, and the detriments were ameliorated by SBF. The neuronal protections of SBF suggest that SBF may have potential benefit in the treatment of neurodegenerative diseases.
