摘  要:目的:用二乙基亚硝胺(DENA)作诱变剂建立SD大鼠肝癌变各时期动物模型,探讨增殖核抗原(PC-NA)及抗酶(AZ)表达的动态变化.方法:从灌胃之日起,在2、4、6、8、10、12、14、16w分8批处死大鼠,取其肝脏,进行病理分级、免疫组化和Western blot,对大鼠肝癌演进中细胞组织学变化及PCNA和AZ的表达进行研究.结果:PCNA随诱癌的进行其表达量显著增加,且从重度炎性反应(HI)至肝胆管细胞癌(CCC)与对照组相比均呈极显著差异(P<0.01);AZ的表达从轻度炎性反应(LI)开始逐渐降低,到非腺瘤样增生(UNH)降至最低值,至CCC时其表达虽略有升高,但仍低于对照组.结论:在SD大鼠肝癌演进中,PCNA的表达逐渐升高,而AZ表达呈降低趋势;两种因子的表达及细胞、组织形态的变化具有一定规律,为研究肝癌的分子机理、临床诊断和治疗奠定重要理论基础.To discuss the dynamic changes of proliferation cell nuclear antigen (PCNA) and antienzyme (AZ) during the hepatocareinogenesis in SD rats, animal models of every pathologic stage are established by n-nitroso-diethylamin (DENA) garage. Animals are sacrificed respectively at 2, 4, 6, 8, 10, 12, 14 and 16 weeks after gavage, and livers are harvested. Then pathological grading, immunohistochemistry and Western blot have been used to explore the cytohistological changes and expressions of PCNA and AZ in the whole process of carcinogenesis. Results show the follwing: There are increased expressions of PCNA with the gavage time, and a significant difference (P〈0.01) compared with the control group from heavy inflammation (HI) to eholangiocellular carcinoma (CCC) ; Expressions of AZ decreas gradually from light infalmmation (LI) and achieve to the lowest at untypical nodus hyperplasia (UNH), then to CCC AZ expression is up-regulated but still lower than the control group. These data suggest that cytohistological changes and expressions of PCNA and AZ may have some regularities. They may provide an important theoretical foundation for studies on molecular mechanism of hepatocarcinogenesis, clinical diagnosis and treatment.
