眼针对大鼠脑缺血再灌注损伤不同时间点脑组织TNF-α表达的影响

摘　　要：目的通过观察不同时限缺血再灌注损伤后脑组织肿瘤坏死因子-α(TNF-α)的表达,探讨TNF-α对脑缺血再灌注损伤的作用机制,并观察眼针对其干预作用。方法 SD大鼠随机分为正常对照组、假手术组、模型组、眼针组;采用改良线栓法复制大脑中动脉缺血再灌注损伤模型,选取眼穴肝区、肾区、上焦区、下焦区,平刺,进针3mm,留针20min;分别于脑缺血再灌注后3、24、72h处死动物并取材;应用免疫组织化学染色法和蛋白质印迹法(Western blot)检测脑组织中TNF-α蛋白表达。结果眼针组TNF-α在脑组织再灌注后3、24、72h均较模型组明显降低,有显著差异。结论眼针疗法可以降低脑缺血再灌注模型大鼠脑组织TNF-α的表达,进而抑制由脑缺血再灌注诱导的炎症反应,发挥脑保护的作用。Objective： To investigate TNF-α expression at different time points in the brain of rats with cerebral ischemia-reperfusion injury,in order to discuss the mechanism and observe the intervention effects of eye-acupuncture therapy.Methods： Healthy SD rats,male and female,were randomly divided into 4 groups： the normal group,the sham-operated control group,the model group and the eye-acupuncture group.The sham-operated control group,the model group and the eye-acupuncture group were divided into the 3h group,the 24h group and the 72h group.The rat model of cerebral ischemia-reperfusion was established by suture method.Eye acupoint liver area,kidney area,upper jiao area and lower jiao area were selected and acupunctured horizontally,inserting needle in 3mm deep,retaining needle for 20 minutes.All animals were executed respectively at 3h,24h,72h after reperfusion and draw materials.The method of immunohistochemistry and Western blot technology was taken to detect the changes of brain TNF-α expression in rats after the eye-acupuncture therapy.Results： Compared with the model group,TNF-α expressions in the brain of rats of the eye-acupuncture group were decreased obviously at 3h,24h,72h after reperfusion.These differences were significant.Conclusion： The eye-acupuncture therapy can regulate down brain TNF-α expressions in rats with cerebral ischemia-reperfusion injury,suppress inflammatory response induced by cerebral ischemia-reperfusion and play a role in protecting brain.
