活血清解汤对大鼠急性胰腺炎早期胰腺细胞凋亡的作用

摘　　要：目的探讨活血清解汤对大鼠急性胰腺炎(AP)治疗作用机制。方法将60只SD雄性大鼠随机分为3组:正常对照组组、非治疗组、活血清解汤治疗组。采用经腹腔注射L-精氨酸制作大鼠急性胰腺炎模型,经注胃管给予生理盐水或中药,每6小时1次。NS组、非治疗组、活血清解汤治疗组分别于造模后24、36h处死各10只。分别取血查血清淀粉酶、TNF、IL-1、IL-10。观察各组胰腺组织的病理变化并评分,同时行末端脱氧核苷酸转换酶介导的原位末端标记检测胰腺细胞凋亡情况。结果活血清解汤治疗组24h、36h血清淀粉酶、TNF、IL-1活性明显下降,而IL-10水平明显上升;胰腺组织出血坏死减轻,24h、36h病理评分均低于非治疗组;给药后24、36h胰腺细胞凋亡指数均高于非治疗组。结论中药活血清解汤能控制过度的炎性反应状态,维持细胞因子网络平衡,诱导非治疗受损胰腺细胞凋亡,起到治疗急性胰腺炎作用。Objective： To study the treatment mechanism of the Huoxueqingjie Decoction on rat model of acute pancreatitis（AP）.Methods： 60 SD male rats were randomly divided into 3 groups with 20 rates in each one including the normal control group,the acute pancreatitis group and the Huoxueqingjie Decoction treatment group.The AP model was made by single intraperitoneal injection of L-arginine.The medicine or normal saline were given by way of gastrotube once every 6 hours.Animals in each group were killed on 24h and 36h after injection.Blood samples were taken for the examination of amylopsin,TNF,IL-1 and IL-10.Pancreatic histopathological score based on microscopic changes was evaluated.Apoptosis of pancreatic cells was detected by terminal deoxynucleotidyltransferase-mediated dUTP-biotionnickend labeling method.Results： Levels including amylopsin,TNF and IL-1 in blood of Huoxueqingjie Decoction group were decreased obviously.However,the level of IL-10 was increased remarkably.The pancreatitis tissue hemorrhagic necrosis was alleviate.Pathologic scores of the Huoxueqingjie Decoction group in the 24h and 36h were both lower than those of the acute pancreatitis group.The apoptopic index of pancreatic cells were increased significantly compared with the AP group at 24 and 36 hours.Conclusion： Huoxueqingjie Decoction can regulate the extreme inflammatory reaction,preserve the balance of the cytokine net and enduce the apoptosis in injured pancreat in AP.
