鸡胚地龙膏对急性软组织损伤模型组织病理学及细胞因子的影响

摘　　要：目的探讨鸡胚地龙膏对动物急性软组织损伤的治疗作用及其机制。方法 105只新西兰大白兔,随机选出21只为正常对照组,剩余84只新西兰白兔制作左后腿部急性软组织损伤模型,造模成功后随机分为模型对照组、阳性药物组(奇正消痛膏组),鸡胚低剂量组和鸡胚高剂量组,每组21只动物。模型对照组不作处理,自然恢复,阳性药物组采用奇正消痛膏治疗(1g/kg),鸡胚低剂量组和鸡胚高剂量组采用鸡胚地龙膏治疗,药物剂量分别为0.15g/kg和0.75g/kg。动态测定新西兰大白兔急性软组织损伤模型组织病理学评分、血浆黏度、组织学及形态计量学指标,同时测定血浆及组织液中IL-1β水平的含量变化。结果鸡胚地龙膏对急性软组织损伤动物模型有良好的恢复作用,鸡胚高剂量组在病情程度评估、组织病理学、血浆及损伤局部组织中白细胞介素-1β的含量下降方面均优于模型组(P<0.05或0.01)。结论鸡胚地龙膏外用治疗新西兰大白兔急性软组织损伤,能明显降低血液黏度、改善微循环和降低炎症介质IL-1β释放,促进损伤组织的修复。Objective To discuss the therapeutic effect and the mechanism of Jipei ointment on animals with acute soft tissue injury. Methods Among 105 New Zealand white rabbits,21 were selected randomly as the normal control group.The other（84） were modeled as acute soft tissue injury to their left hind legs.After the success of modeling,these rabbits with acute soft tissue injury were divided into four groups：the model control group,the positive drug group（the Qizheng pain relieving ointment group）,the low dose Jipei ointment group and the high dose Jipei ointment group,with 21 rabbits in each group.The model control group was not treated by medication and they were left recovered naturally.The positive drug group was treated by Qizheng pain relieving ointment,while the low dose Jipei ointment group and the high dose Jipei ointment group were treated by Jipei ointment,with 0.15 g/kg and 0.75 g/kg as doses respectively.Histopathological score,plasma viscosity,histology and morphometric index of the model of New Zealand white rabbits with acute soft tissue injury were assayed dynamically.Meanwhile the changes of content of IL-1β level in plasma and tissue fluid were assayed and were analyzed statistically. Results Jipei ointment has good effect on recovery of the model of animals with acute soft tissue injury.The high dose Jipei ointment group is better than the model group in terms of assessment of illness severity,histopathology,decreased content of IL-1β in plasma and damaged local tissue（P〈0.05 or 0.01）. Conclusion Topical application of Jipei ointment on New Zealand white rabbits with acute soft tissue injury can significantly reduce blood viscosity,improve the microcirculation,reduce the release of inflammatory mediator（IL-1β） and accelerate the repair of damaged tissue.
