摘  要:目的建立测定人血浆中氢氯噻嗪质量浓度的液相色谱-串联质谱法。方法以磺胺甲基异唑为内标,采用Agilent ZobraxSB C18柱(100 mm×2.1 mm,3.0μm)为固定相,甲醇-水(80∶20,V/V)为流动相,流速为0.15 mL/min;质谱采用电喷雾离子源,多反应离子监测模式,氢氯噻嗪和内标的检测离子对分别为m/z 295.7→268.7和m/z 251.8→156.0。结果血浆中氢氯噻嗪质量浓度在1.070~214.4μg/L范围内与峰面积比值线性关系良好(r2=0.998 8),最低定量浓度为1.070μg/L,日内、日间精密度的RSD均小于13.9%,相对回收率均在100.5%~102.2%范围内。结论该方法具有较高的准确度,线性范围宽,方法灵敏、专一性好、操作简便,适用于氢氯噻嗪血药浓度的测定及临床药代动力学研究。Objective To determine the concentration of hydrochlorothiazide in human plasma by LC-MS/MS method.Methods Sulfamethoxazole was used internal standard.The evaluation of hydrochlorothiazide was carried out by Agilent Zorbax SB column(100 mm ×2.1 mm,3.0 μm) with the mobile phase of methanol/water(80 ∶20,V/V),and the ions of analyte were detected by multiple ionic reaction monitoring mode(MRM),the mass transition pairs of m/z 295.7→268.7 and m/z 251.8→156.0 were used to detect hydrochlorothiazide and IS,respectively.The flow rate was 0.15 mL/min.Results The calibration curve for plasma hydrochlorothiazide was linear in the range of 1.070-214.4 μg/L(r=0.998 8).The detection limit of hydrochlorothiazide was 1.070 μg/L.Within-day RSD and between-day RSD were less than 13.9%,the relative recovery were all within 100.5%-102.2%.Conclusion This method is accurate,sensitive,specific and convenient,which could be used in the determination of plasma hydrochlorothiazide and its pharmacokinetic study in clinic.
