摘  要:为解决黄心春结球白菜小孢子胚诱导难的问题,以引自韩国和日本的12个黄心春结球白菜杂交种为试材,对影响小孢子胚发生和发育的因素进行了研究。结果表明:供试品种间小孢子胚发生能力有极显著差异;不同品种对培养基中添加植物生长调节剂的反应不同;发现了花蕾低温密封预处理具有促进小孢子成胚的作用;MS培养基中添加0.1mg.L-1的NAA具有促进小孢子植株生根的作用。在优化培养条件的基础上,建立了针对黄心春结球白菜的游离小孢子高效培养体系,获得了11个黄心春结球白菜杂交种的445株小孢子植株,鉴定出362株双单倍体。Isolated microspore culture was carried out with 12 F1 yellow Chinese cabbage introduced from South Korea and Japan.Frequencies of embryo-genesis,factors influencing plantlet regeneration were studied.The results showed that genotypes had very significant effect on frequency of embryogenesis.The reaction of genotypes to growth regulators added in medium was different.Low temperature and hermetic sealing could promote the development of embryoids.The optimal medium for rooting was MS +0.1mg·L-1 NAA.An efficient culture system for yellow heading spring Chinese cabbage was established through the optimization of culture conditions,and totally 445 regenerated plants of 11 hybrid cultivar were obtained,in which 362 were identified as double haploid.
