摘 要:目的研究不同浓度姜黄素(Cur)对正常血管内皮细胞活力的影响,及Cur对血管内皮细胞过氧化氢(H2O2)损伤的保护作用及机制。方法用不同浓度Cur(12.5μmol/L、25μmol/L、50μmol/L、100μmol/L)处理体外培养的人脐静脉内皮细胞(HUVECs)8 h,MTT法检测不同浓度Cur对HUVECs存活率的影响;H2O2(250μmol/L)孵育建立HUVECs损伤模型,不同浓度Cur(12.5μmol/L、25μmol/L、50μmol/L、100μmol/L)与H2O2共同处理HUVECs 4 h,检测细胞存活率,黏附能力和培养液中乳酸脱氢酶(LDH)释放量;Griess法测定培养液中NO2-浓度,反映内皮细胞释放一氧化氮(NO)量。结果不同浓度Cur处理内皮细胞8 h后,内皮细胞的存活率无统计学差异(P>0.05);Cur能明显提高H2O2损伤的内皮细胞存活率和黏附能力(P<0.01),降低内皮细胞LDH的释放(P<0.01);增加H2O2损伤后HUVECs培养液中NO含量(P<0.01)。其中Cur浓度为25μmol/L时效果最明显。结论不同浓度Cur(12.5μmol/L、25μmol/L、50μmol/L、100μmol/L)对内皮细胞的存活率均无影响;Cur对血管内皮细胞氧化损伤具有保护作用,机制可能与其促进NO的合成有关。Objective To study the effects of curcumin on normal vascular endothelial cells, and the protective effects and mechanism of curcumin against H2O2 - induced vascular endothelial cells injury. Methods The cultured human umbilical vein endothelial cells (HUVECs) were treated with Cur (12.5 μmol/L, 25 μmol/L, 50 μmoL/L, 100 μmol/L) for 8 h, the survival ratio of HUVECs was detected with MTT assay. The injury of HUVECs was induced by H2O2 (250 μmol/L) treatment, and varies concentration of Cur ( 12.5 μmol/L, 25 μmol/L, 50 μmol/L, 100μmol/L) were applied together with H2O2 for 4 h. The survival ratios, the adhesion capacity, the LDH releasing and the NO2- concentrations of HUVECs were detected. Results After treatment with curcumin ( 12.5 μmol/L, 25 μmol/L, 50 μmol/L, 100 μmol/L) for 8 h, the survival ratio of HUVECs did not change significantly ( P 〉 0.05). The survival ratio and adhesion capacity of H202 - treated HUVECs were improved and the LDH releasing was reduced significantly by curcumin treatment ( P 〈 0.01 ). In addition, curcumin treatment significantly increased the production of NO in culture medium com- pared with n2 02 treatment alone ( P 〈 0.01 ). These protective effects were much more significant than when concentration of cur was 25 μmol/L compared with other concentrations observed. Conclusion Different concentrations of curcumin (12.5 μmol/L, 25 μmol/ L, 50 μmol/L, 100 μmol/L) observed in this study have no significant effects on the survival ratio of HUVECs, which implies that the safe concentration range of curcunin is wild. Curcumin treatment can protect against H2O2 induced injuries on cultured vascular endothelial cells via its antioxidant capacity and NO synthesis promotion.
