摘 要:观察汉族人群肝脂肪酶(HL)基因2个标签单核苷酸多态性(SNP)与2型糖尿病(T2DM)及血脂代谢的相关性。应用荧光标记单碱基延伸分型技术及寡核苷酸微阵列芯片杂交技术检测HL基因rs12462668和rs10426971多态性。用全自动生化分析仪检测CHO、TG、HDL-C、LDL-C、载脂蛋白A1(ApoA1)及载脂蛋白B(ApoB)。结果表明:T2DM组rs12462668的AA、GG基因型频率、等位基因频率及由2个SNP构建的单倍型频率与对照组比较差异有统计学意义。T2DM组的TG和ApoB显著高于对照组,HDL-C和ApoA1显著低于对照组。组间各基因型血脂水平比较结果显示,2个SNP与TG、HDL-C和ApoA1代谢紊乱有关。组内各基因型间血脂水平比较结果显示,T2DM患者的HDL-C水平与rs12462668多态性有关,而TG水平与rs10426971多态性有关。这提示HL基因的2个标签SNP与T2DM的发生密切相关,并可影响血脂的代谢。To explore the association between 2 tag single nucleotide polymorphisms(SNP) in hepatic lipase(HL) gene and serum lipid levels in type 2 diabetes mellitus(T2DM) in Chinese Han nationality.Rs12462668 and rs10426971 of HL gene were analyzed with fluorogenic single nucleotide extension genotype and oligonucleotide microarray hybridization.The serum levels of CHO,TG,HDL-C,LDL-C,Apo A1 and Apo B were detected by automatic biochemical analyzer.The results showed that the frequencies of AA,GG genotype,allele of rs12462668 and haplotype constructed with the two tag SNPs were differ between the two groups.The serum levels of TG and Apo B were higher,HDL-C and Apo A1 were lower in T2DM group than that in control group.The serum lipid results between two groups showed that genotype of the 2 SNPs were associated with disorder metabolism of TG,HDL-C and Apo A1.When serum lipid levels were compared among different genotypes in each group,it showed that HDL-C levels were associated with rs12462668a and TG levels were associated with rs10426971 in T2DM group.The results suggest that two tag SNPs of HL gene are related to T2DM and may influence lipid metabolism.
