摘 要:目的研究环境雌激素壬基酚(nonylphenol,NP)暴露对雌性SD大鼠脾组织及脾淋巴细胞的损伤作用,探讨壬基酚对雌性大鼠免疫功能的影响。方法将32只健康8~9周龄SPF级雌性SD大鼠按体重随机分为溶剂对照组(玉米油)和低(20 mg/kg)、中(80 mg/kg)、高(200 mg/kg)剂量壬基酚染毒组,每组8只。采用灌胃方式进行染毒,灌胃容量5 ml/kg,隔天染毒1次,连续60 d。观察脾组织病理学改变。采用流式细胞仪检测脾淋巴细胞内活性氧(ROS)、游离钙离子([Ca2+]i)浓度、线粒体膜电位(MMP)水平的变化。结果与溶剂对照组相比,中、高剂量壬基酚染毒组大鼠脾淋巴细胞内ROS水平和高剂量壬基酚染毒组脾淋巴细胞内[Ca2+]i水平明显升高,高剂量壬基酚染毒组脾淋巴细胞内MMP水平明显降低,差异均有统计学意义(P<0.05,P<0.01)。且脾淋巴细胞内ROS[、Ca2+]i水平与壬基酚浓度呈正相关(ROSr:=0.488,P<0.05[;Ca2+]ir:=0.497,P<0.05),MMP水平与壬基酚浓度呈负相关(r=-0.565,P<0.05)。病理学检查可见,中、高剂量壬基酚染毒组脾脏白髓减少,脾窦内血管扩张充血。结论壬基酚对雌性大鼠脾脏及脾淋巴细胞具有一定的毒性作用,并可引起免疫功能损伤。Objective To study the adverse effects of nonylphenol (NP) on spleen tissue and splenocytes of female rats, and investigate the effects of NP on immune function of female rats. Methods Thirty-two female SD rats were randomly divided into four groups,eight in each,treated with NP through gavage at the doses of 0,20,80 and 200 mg/kg respectively, once two days for 60 consecutive days. In the end of the experiment,the pathological examination of the spleen was done and the level of reactive oxygen species (ROS), intracellular free calcium concentration ([Ca2+]i) and mitochondrial membrane potential (MMP) in spleen lymphocytes were detected by flow cytometry. Results Compared with the control, the levels of ROS and calcium in spleen lymphocytes increased dramatically in the 200 mg/kg NP treated group. Both of them were in a dosedependent manner (ROS: r=0.488, P〈0.05; [Ca2+]i:r=0.497, P〈0.05). NP induced reduction of the level of MMP and negative relation between the level and dose was found (r=-0.565 ,P〈0.05). Histopathological changes were found among the spleen of the experimental groups that characterized with the white pulp reduced, splenic corpuscle shrunk and spleen sinus vascular dilatation and congestion. Conclusion Oral exposure to nonylphenol may induce adverse effects on the spleen and spleen lymphocyte in rats and 'affect the immune function of female rats.
