摘 要:目的探讨深低温冻储对大鼠同种异体移植气管上皮再生的影响。方法用深低温冻储2周和6周的大鼠气管进行同种异体气管移植,以自体和异体新鲜气管作为对照,通过观察供体气管的组织学、上皮爬行和再生情况,评价深低温冻储不同时长对移植气管的存活和新生上皮再生的作用。结果深低温冻储6周后实验组的移植气管复层上皮比自体气管移植组厚度明显变薄或成单层,软骨无变性坏死,未见有单核细胞浸润,最长存活达35 d,而深低温冻储2周气管移植大鼠为10 d。结论深低温冷冻后的组织可以保持其原有的活力和组织完整性,并能消减供体气管的抗原性。冻储6周的移植气管上皮再生和受体的存活率要优于冻储2周组。Objective To investigagte the effect of cryopreservation on tracheal allograft epithelium regereneation.Methods After transplanted with cryopreserved 2 weeks and 6 weeks allocraft,the histology of the transplanted trachea including epithelium growth and regeneration was observed to evaluate the the effect of cryopreservation on tracheal allograft epithelium regereneation.Results The implant trachea of ALLO-6wk rats had similar structure as that of AUTO-control group.There were no monocyte infiltration.Epithelium regeneration in the ALLO-6wk group was better than that in the ALLO-2wk group.The longest survival time in ALLO-6wk group was 35 d,compared with 10 d in ALLO-2wk group.Conclusion Cryoperserved allografts retained their vitality and intact structure.Trachea that were cryopreserved for 6 weeks had much lower immunogenerocity than those were cryopreserved for 2 weeks.
