摘 要:目的:探讨亲代大鼠配对前经历吗啡成瘾及戒断对子代掠食行为的影响及机制。方法:选用8周龄Sprague-Dawley大鼠40只建立吗啡依赖模型,自然戒断21 d后配对分组合笼,子鼠8周龄时进行掠食行为观察,采用Golgi-Cox染色观察子鼠前扣带皮质内神经元树突形态情况。结果:吗啡成瘾组子代掠食量均低于对照组(P<0.001);吗啡成瘾组子代前扣带皮质内神经元顶树突分枝数、分枝总长度和树突棘密度均低于对照组(P<0.05)。结论:亲代经历吗啡成瘾戒断会导致子代掠食行为障碍,其机制可能与前扣带皮质内神经元树突发育不良有关。Objective:To investigate the effects and possible mechanisms of morphine addiction and withdrawal in parents prior to mating on the foraging behavior in adult offspring rats.Methods:Eight-week healthy adult Sprague-Dawleyda(SD) rats were used to eatabilish morphine-addiction model(n=40).Male and female rats were mated 21 days after morphine withdrawal.The foraging behavior tests were assessed when the offsprings were 8 weeks old.Golgi staining was conducted in order to analyze the neuronal morphology of anterior cingulate cortex.Results:Much fewer food was foraged by offsprings of morphine-addicted group compared with control group(P0.001);the numbers of apical dendrites branches,the total length of apical dendrites branch and apical dendritic spine density in anterior cingulate cortex neurons of offsprings from morphine-addicted group were all lower than those from control group(P0.05).Conclusion:The experience of morphine addiction and withdrawal prior to mating will lead to food foraging function deficit in the offspring,which might be related to dysplasia of dendritic morphology in anterior cingulate cortex neurons.
