摘 要:目的:探讨羟基喜树碱(HCPT)对体外培养的人胚肺成纤维细胞(HFL1)增殖与凋亡的影响。方法:常规培养HFL1细胞,以未加HCPT的HFL1为对照组,以不同浓度梯度(0.008~32 mg/L)HCPT处理的HFL1作为实验组,采用MTT法检测HCPT干预24、48、72 h后HFL1的增殖情况;光学显微镜观察干预后细胞的形态学变化;流式细胞仪检测细胞凋亡率;透射电镜下观察细胞凋亡的改变。结果:HCPT对HFL1有明显的抑制作用,在一定程度上呈现剂量-时间依赖性;HCPT处理组HFL1凋亡率较阴性对照组显著升高(F=399.51,P<0.01),并呈药物浓度依赖性。HCPT 0.5 mg/L作用HFL1 48 h,透射电镜下可见细胞缩小,核仁消失,染色质浓聚的凋亡状态。结论:HCPT在体外通过抑制细胞增殖和促进细胞凋亡来抑制HFL1的生长。Objectives To investigate the effects of hydrocamptothecin (HCPT) on the proliferation and apoptosis of human fetal lung fibroblasts cell line HFL1 in vitro. Methods Human fetal lung fibroblasts cell line HFL1 were cultured in vitro, and exposed to HCPT with different final concentrations (0.008-32 mg/L respectively), cells without HCPT exposure served as the control. The cell proliferation was assessed by MTF colofimetric assay. The morphological change of the HFL1 were observed with light microscopy. The rate of cell apoptosis was identified by FCM. Results MTF assay indicated that HCPT significantly inhibited the proliferation of HFL1 in a dose-and time-dependent manner, apoptosis rates were significantly higher than that of control group (P 〈 0.01). After treatment with the concentration of 0.5 mg/L for 48 h, typical apoptotic morphological features of cell shrinkage, nucleoli disappearance and condensation and margination of nuclear chromatin were observed by TEM. Conclusion HCPT could significantly inhibit the proliferation of HFL1 in a dose-and time-dependent manner, and could induce apoptosis of HFL1 in vitro.
