摘 要:目的:观察苦参碱对受192铱(192Ir)低剂量照射人员骨髓细胞端粒酶活性及细胞凋亡的影响。方法:以受192Ir迁延性低剂量照射(0.05~0.66 Gy)患者54例为研究对象,分别予复方皂矾丸、苦参碱和安慰剂组治疗。采用端粒重复扩增(TRAP)-ELISA方法检测端粒酶活性,AnnexinV-FITC及PI双染,应用FACScan进行流式细胞术定量检测细胞凋亡率,观察受照人员治疗前后骨髓象、骨髓细胞端粒酶活性、细胞凋亡率变化。结果:受照人员骨髓各系均可见形态学异常,总异常率(减低+极度活跃)为20.4%;骨髓细胞端粒酶活性随时间迁延而升高,细胞凋亡率则随时间迁延而降低。经苦参碱及复方皂矾丸组治疗后6个月各系形态异常、端粒酶活性亦逐步恢复接近正常,骨髓细胞凋亡率增高。其中以苦参碱组治疗后疗效更为显著(P<0.01)。结论:苦参碱作为辐射防护剂可诱导受辐照射人员骨髓细胞凋亡,抑制其端粒酶活性高表达。Objective To observe the effect of matrine on telomerase activity & apoptosis of bone marrow cells in humans exposure to 192Ir low-dose radiation. Methods Extent of bone marrow cell proliferation were observed by smears; Telomerase activity of bone marrow cell were detected by TRAP-ELISA (telomerie repeat amplification protocol-Enzyme linked immunosorbent assay). Apoptosis rate were detected by flow cytometry (FCM) after marked with Annexin V-FITC and PI. A study was carried out on effects of matrine, fu fang zao fan pills, and placebo as treatments , and observe the proliferation, telomerase activity and apoptosis of bone marrow cells in humans exposure to 192Ir low-dose radiation (54 men,0.05 - 0.66 Gy). Results Cell morphous of all lines were abnormal in irradiated people,The total abnormal rate is up to 20.4% including hypoplasia and polarhypoplasia; The telomerase activity of bone marrow cell was obviously higher than normal with the time go in irradiated group, The apoptosis rate of irradiated group was degrade with the time go; Cell morphous of all lines was gradually up to normal by treat with matrine and fu fang zao fan pills, so as the telomerase activity,and the apoptosis rate wasincreased,the effect of matrine treatment group was significantly better than others (P 〈 0.01 ). Conclusion As a protective agent from irradiation, matrine could surppress telomerase activity in men irradiated by 192Ir, and induce apoptosis of bone marrow.
