摘 要:目的探讨感染泡球蚴的小鼠皮肤移植后免疫系统的变化对移植物的影响。方法采用小鼠尾部皮肤移植建立小鼠皮肤移植模型。将64只C57BL/6小鼠及64只BALB/c小鼠各随机分为4组,每组各16对小鼠,其中16只C57BL/6小鼠为供体,16只BALB/c小鼠为受体。A组:空白对照组(非同系移植不予以任何干预);B组:实验组1,以健康C57BL/6小鼠尾部皮肤移植至BALB/c小鼠(感染泡球蚴1个月后);C组:药物对照组,采用非同系移植加药物[西罗莫司(商品名:雷铂霉素)]干预,皮肤移植术后5d每日给予西罗莫司1.5mg/kg干预;D组:实验组2,以健康C57BL/6小鼠尾部皮肤移植至BALB/c小鼠(感染泡球蚴3个月后)。术后观察BALB/c小鼠尾部移植皮肤生长情况。每组留8只用于观察BALB/c小鼠术后尾部移植皮肤生存期,每组于术后3、5、7、9d4个时间点各取2只小鼠尾部移植皮片,用于观察小鼠尾部移植皮肤病理变化情况。结果 A、B、C、D组移植皮肤存活时间平均为(6.00±0.76)、(7.50±0.93)、(13.25±1.04)、(11.5±0.93)d。C组小鼠移植皮片存活时间较D组长,差异有统计学意义(P<0.05)。C、D组移植皮肤存活时间较A、B组延长,差异有统计学意义(P<0.01)。组织病理学检查:A、B组移植皮片均在术后1周左右发生明显的逐渐加重的急性免疫排斥反应,C、D组移植皮片的免疫排斥反应较A、B组轻。结论小鼠感染泡状棘球蚴后有利于同种异体移植皮片的存活,可以延长移植皮片的生存时间。该模型为研究感染泡球的免疫实验研究提供了良好的实验平台,为进一步研究泡球蚴感染后机体免疫状态的变化提供了理论依据。Objective To investigate the immune characteristics of mice infected with Echinococcus Multilocularis(E.m) and its effect on the graft by using tail skin transplantation models.Methods Tail skin transplantation method was adopted in the C57BL/6-to-BALB/c mice skin grafting models.64 C57BL/6 mice and 64 BALB/c mice were randomly divided into 4 groups as follows,16 pairs of mice in each group,16 C57BL/6 mice as donator,16 BALB/c mice as acceptor: Group A: normal controls(tail skin transplantation in the different strain without any intervention);Group B: healthy C57BL/6 mice to BALB/c mice with E.m infection for one months.Group C: medicine controls(tail skin transplantation in the different strain with medicine intervention: neotype immune depressant Sirolimus): Sirolimus(1.5 mg/kg) was used after skin transplantation for five days.Group D: healthy C57BL/6 mice to BALB/c mice with E.m infection for 3 months.Observe survival condition of transplanted skin on mice tail after operation.Eight BALB/c mouse in each group were remained for observation of their survival time of transplanted skin on tail.The transplanted skin samples were collected in 3,5,7,9 days after operation respectively to observe pathological change of transplanted skin.Results The mean survival times of allograft in A,B,C and D group were(6.00±0.76)d,(7.50±0.93)d,(13.25±1.04) d,and(11.5±0.93)d,respectively.The allograft survival times of group C and D were longer than that of group A and B.There were statistical significant(P0.01).The pathological results indicated: The allograft in group A and B showed conspicuous acute immunological rejection in one week after operation.The immunological rejection of allograft in group C and D were weaker than group A and B.Conclusion E.m infection was beneficial to the survive of allograft after operation,could prolong the survival time of allograft.This model was a good platform for further research in immune empirical study after E.m infection,establish f
