摘 要:目的探讨应激对大鼠红细胞免疫功能的损伤机制。方法雄性Wistar成年大鼠20只,随机分为应激模型组和正常对照组。建立应激动物模型,应用放射免疫分析法观察大鼠血浆皮质酮的变化,应用酵母菌花环法测定红细胞各花环率,采用黄嘌啉氧化酶法测定大鼠红细胞膜超氧化物歧化酶(Erythrocyte-Superoxide dis-mutase,E-SOD)活性,应用硫代巴比妥法测定脂质过氧化物(Erythrocyte-Malondialdehyde,E-MDA)含量。结果模型组大鼠肝脏、大脑、心脏、脾脏、肾脏系数与对照组比较差异有统计学意义(P<0.05),应激第7、14d血浆皮质酮水平高于对照组(P<0.05);红细胞C3b受体花环率低于对照组(P<0.05),红细胞免疫复合物花环率高于对照组,但无明显差异(P>0.05);模型组E-SOD活性低于对照组(P<0.05),E-MDA含量增高(P<0.05);且红细胞C3b受体花环率与E-SOD活性、E-MDA含量存在相关性。结论应激诱发大鼠红细胞免疫功能多项指标改变,且应激时皮质酮的升高、自由基堆积所致脂质过氧化损伤可能均是影响红细胞免疫功能的重要原因。Objective To study the effects of stress on erythrocyte immune functions in rats and explore mechanism of it. Methods The rat stress model was established, and levels of cortisol in blood were measured. And then Erythrocyte Immune Function was measured by the method of Yeast Rosette, the activities of Erythrocyte-Superoxide dismutase (E-SOD), the contents of Erythrocyte -Malondialdehyde (E-MDA) were measured and analyzed respectively with xanthine oxidase and thiobarbituric acid methods in two groups. Results Rats, liver, brain, heart, spleen, kidneys coefficient compared with the control group was significantly (P 〈0.05), section 7,14 d stress plasma corticosterone levels higher (P 〈0.05); C3b receptor rosette rate of red blood cells than the control group (P 〈0.05), red cell immune complex rosette rate was higher, but no significant difference (P 〉 0.05); model group, E-SOD activity was lower than the control group (P 〈0.05), E-MDA levels increased (P 〈0.05); and the C3b receptor rosette rate of red blood cells and E-SOD activity, E-MDA levels correlated. Conclusions The repeated stress can influence the erythrocyte immune system which maybe the reason of the hormone increased in blood and the abnormal metabolism of free radicals causing lipid peroxidation.
