糖尿病兔后肢缺血模型的制作

摘　　要：目的探讨建立糖尿病兔后肢缺血模型的制作方法。方法新西兰大耳白兔30只,四氧嘧啶(alloxan,ALX)静脉注射建立糖尿病兔模型。建模成功后,结扎并离断糖尿病兔右后肢的髂动脉远端并切除股总和股浅动脉导致缺血。术后普食喂养,定期评估家兔缺血程度,直至术后8周。结果 30只兔参与建模,4只死于ALX给药后,26只兔糖尿病模型建模成功。血管造影显示术后大腿的细微侧支血管记数率从0.56(s=0.21)增加到1.54(s=0.26)。右后肢肌肉的组织学研究显示有萎缩、坏死。结论成功制作糖尿病兔后肢缺血的实验模型,模型稳定、重复性好。Objective To establish an experimental animal model of limb ischemia in diabetic rabbits. Methods The diabetic model was established by administration of alloxan in 30 rabbits. Ischemia was induced by ligation of the distal iliac artery and excision of the common and superficial femoral arteries in the right hindlimb of 26 rabbits with stable diabetes. All the rabbits were given normal feeding. The severity of the ischemia in each animal was evaluated regularly until 8 weeks after the surgery. Results A total of 30 rabbits were modeled, 4 of them died after administration of ALX, 26 of them were modeled successfully. The postoperative angiography showed that the count rate of subtle collateral vessels increased from 0.56 （ s = 0.21 ） to 1.54 （ s = 0.26）. The histological study of the hindlimb muscle showed atrophy, fibrosis and necrosis. Conclusions Persistent hindlimb ischemia model in the diabetic rabbit is established successfully, and the model is stable and repeatable.
