益气解毒法不同组方对鼻咽癌细胞迁徙运动潜能及细胞伪足的影响

摘　　要：目的研究益气解毒法不同组方对鼻咽癌细胞迁徙运动潜能及其细胞伪足的影响。方法以正常大鼠血浆为对照,分别应用益气解毒方原方及其改方大鼠药物血浆对CNE2Z细胞进行干预,采用划痕实验法评价不同组方药物对靶细胞迁徙运动潜能的影响,以扫描电镜观察细胞伪足形态特征,分析靶细胞的反应特性。结果益气解毒原方对CNE2Z细胞的迁徒运动潜能显示抑制效应,细胞迁徙能力较差,伪足伸展不明显;益气解毒改方则对靶细胞迁徙运动潜能显示为促进效应,细胞伪足伸展长度较长。结论同为益气解毒法的组方,由于剂量构成比不同,各方对鼻咽癌细胞迁徙运动潜能的药效学差异巨大。这样的药理效应特点,对中药复方的合理组方提出了严格要求。Objective To investigate the effects of original and modified Qi-Boosting Toxin-Resolving Formulae （QBTRF） on moving potentiality and cellular pseudopod of nasopharyngeal carcinoma cell line CNE2Z based on an experimental study. Methods Investigated were the effects of two herbal medicinal formulae, the original and modified QBTRF, on moving potentiality and cellular pseudopod of NPC cell line CNE2Z by medicinal blood plasma assaying procedures, with healthy rat plasma as the control in a routine way. Here, scratch test was taken to determine the moving potentiality with their changes in morphologic characteristics of cells CNE2Z under light microscope, and electron microscopy was utilized to observe pseudopod features of these cells to evaluate their changes in potential motility in the response to the intervening reagents. Results It was sho^vn that the original QBTRF brought about an inhibitory effect on the moving potentiality of CNE2Z cells showing a much shortened moving distance with looser growing status and silkiness- like pseudopod expanding not so obvious as that in controlling cells, while the modified OBTRF brought about a promoting effect on the moving potentiality of CNE2Z cells showing a much strengthened moving ability showing a very longer moving distance with closer growing status and silkiness-like pseudopod expanding much longer than that in controlling cells. Conclusions Though the original QBTRF and the modified one are both composed of the same therapeutic principles, i.e. Qi-boosting and toxin-resolving, they hold much different pharmacodynamics on the moving potentiality and cellular pseudopod of CNE2Z cells as a result of changes in dosage ratio composing of some herbs, Such a characteristic suggests that a highly reasonable composing principle for a compound herbal formula should be a very important for ideal therapeutic effect obtained in clinical practice.
