外周血TREM-1在老年SIRS患者中的表达及其意义

摘　　要：目的检测外周血髓样细胞触发性受体1-(TREM-1)mRNA的表达水平,探讨其在老年全身炎症反应综合征(SIRS)患者中的表达及其意义。方法采用SYBRGreenI实时荧光定量RT-PCR技术,以β-actin为内参照,检测22例老年SIRS患者〔SIRS组,其中多器官功能障碍综合征(MODS组)6例〕和23例健康者(对照组)外周血TREM-1mRNA表达水平。结果 TREM-1和β-actin的熔解曲线均仅有单一峰。SIRS组Ct值(β-actin/TREM-1)为0.930±0.066,正常对照组Ct值为0.846±0.021,2组比较差异具有统计学意义(P<0.01);MODS组Ct值为1.006±0.037,与SIRS组比较差异亦有统计学意义(P<0.05),显著高于正常对照组(P<0.01)。结论应用RT-PCR技术检测TREM-1在外周血中的表达水平简便易行,结果稳定可靠。TREM-1可作为SIRS、MODS早期诊断和病情判断的指标,可能成为SIRS、MODS一个潜在的治疗靶点。Objective To detect the expression of triggering receptor expressed on myeloid ceils -1 （TREM-1） mRNA in human peripheral blood by SYBR Green I reverse transcription polymerase chain reaction （RT-PCR） and explore the ex- pression and significance of TREM-1 of elder systemic inflammatory response syndrome（SIRS） patients. Methods Real-time fluorescent quantitative RT-PCR was used in our test to investigate the expression of TREM-1 mRNA in human peripheral blood with β-actin as the internal control gene. Total 22 elder SIRS patients and 23 healthy were involved. Results The analysis of dissociation curves on TREM-1 and β-actin showed that both of them had only one simple spike. The values of β-actin/ TREM-1 （Ct） in control group was 0.846±0.021 ,The values of Ct in SIRS group was 0.930 ± 0. 066. The statistical difference was significant between control group and SIRS group （ P 〈 0.01 ）. The values Ct in MODS group was 1. 006 ±0. 037, which was higher than that in control group apparently（ P 〈 0. 01 ）. There was a statistical difference between SIRS group and MODS group（ P 〈 0.05 ） also. Conclusions It is an easy and reliable test to detect the expression level of TREM-1 in peripheral blood by SYBR Green I real-time fluorescence quantitative RT-PCR. Expressions of TREM-1 among the SIRS, MODS and healthy person have significant difference. TREM-1 can be used as an index in the early diagnosis and illness judgement. It may become a potential therapeutic target.
