摘 要:目的探讨穿心莲有效成分(API0134)是否具有抑制小鼠腹腔巨噬细胞基质金属蛋白酶-2和基质金属蛋白酶-9(MMP-2,MMP-9)活性及其mRNA表达的作用。方法用腹腔冲洗的方法收集小鼠腹腔巨噬细胞,使用氧化低密度脂蛋白(ox-LDL)刺激巨噬细胞,同时分别以不同浓度的API0134对巨噬细胞进行干预。采用免疫荧光方法、明胶酶谱法和RT-PCR的方法检测ox-LDL和API0134对巨噬细胞MMP-2、MMP-9活性和表达的影响。结果与对照组相比,ox-LDL组MMP-2及MMP-9活性和mRNA表达显著升高(均P<0.01)。与ox-LDL组相比,加入50μg/mL API0134组MMP-2及MMP-9的活性和mRNA表达变化不明显(均P>0.05),而加入100、200μg/mL API0134组的MMP-2及MMP-9的活性和mRNA表达均有不同程度的下降(均P<0.05)。结论穿心莲有效成分能够抑制ox-LDL对巨噬细胞MMP-2及MMP-9活性和mRNA表达的上调作用,因此可能具有维护粥样斑块稳定,减少斑块破裂危险的作用。Objective To observe the effects of Andrographis paniculata active ingredient API0134 on activity and expression of matrix metalloproteinase(MMP)-2 and MMP-9 in murine peritoneal macrophages.Methods The macrophages were isolated in the peritoneal lavage of mice,and stimulated with ox-LDL(100 μg/mL)and intervened with API0134 at different concentrations respectively.By using the immunofluorescence,the Gelatinase zymography and the RT-PCR method the changes in activity and expression of MMP-2 and MMP-9 in macrophages were observed.Results As compared with the control group,the activity and expression of MMP-2 and MMP-9 in ox-LDL group were obviously elevated(all P0.01),and there was no significant difference in the change of activity and expression of MMP-2 and MMP-9 between 50 μg/mL API0134-treated group and ox-LDL group(all P0.05),but the expression levels of MMP-2 and MMP-9 in 100 and 200 μg/mL API0134-treated groups were significantly decreased as compared with ox-LDL group(all P0.05).Conclusion API0134 can inhibit the activity and expression of MMP-2 and MMP-9 in macrophages,suggesting it may maintain the stability of plaque and reduce the risk of plaque rapture.
