摘  要:目的探讨经典瞬时受体电位(transient receptor potential canonical,TRPC)通道在慢性哮喘小鼠肺组织中的表达。方法随机将BALB/c小鼠分成对照组和哮喘组。应用5%鸡卵白蛋白(OVA)腹腔注射致敏和雾化激化的方法制备小鼠慢性哮喘模型,对照组用生理盐水代替OVA。最后一次激发24h后处死小鼠,取肺组织进行RT-PCR和免疫组织化学检测,观察哮喘小鼠肺组织TRPC mRNA和蛋白表达水平的变化。结果 RT-PCR结果显示,正常小鼠肺组织分别表达TRPC1、TRPC2、TRPC3、TRPC6mRNA;与对照组相比,哮喘小鼠肺组织TRPC1、TRPC6mRNA表达显著增高(P<0.05);免疫组织化学检测发现TRPC1、6蛋白在哮喘小鼠肺组织的支气管上皮细胞、血管平滑肌细胞及浸润的炎症细胞表达明显增强。结论 TRPC1、TRPC6可能与哮喘发病机制密切相关。Objective To study the mRNA and protein expression of canonical transient receptor potential protein(TRPC)in lung tissues of mice with chronic asthma.Methods BALB/c mice were randomly divided into two groups:control group and asthma model group.The mice in the asthma model group were sensitized with 5% OVA by intraperitoneal injection and subsequently challenged by OVA inhalation,while those in control group were similarly treated with sterile saline.Mice were sacrificed at 24th h after the last OVA challenge.The mRNA and protein expression in lung tissues was detected by using RT-PCR and immunohistochemistry.Results The mRNA expression of TRPC1,TRPC2,TRPC3 and TRPC6 was detected in normal lung tissues.As compared with the control group,the mRNA level of TRPC1 and TRPC6 in the asthma model group was increased(P0.05).The TRPC1 and TRPC6 protein levels in the mice with asthma were increased in bronchial epithelial cells,vascular smooth muscle cells and inflammatory cells.Conclusion TRPC1 and TRPC6 are involved in asthma.
