家兔膝关节创伤性骨化性肌炎动物模型的建立

摘　　要：目的　建立家兔膝关节创伤性骨化性肌炎动物模型。方法　用新西兰白色家兔10只,进行强力被动扭转、暴力按摩,造成膝关节及周围软组织损伤,然后固定膝关节于伸直位。从第1周开始每周按摩6天,第5周开始改为隔天按摩一次,并观察皮温、肿胀度、局部体征、局部骨化情况。至第9周进行病理、组织学检查。结果　模型局部肿胀明显;皮温升高;患肢关节强直,伸屈功能明显障碍,最后关节僵硬,膝关节周围肌群可触摸到肌肉硬块;X线片示全部出现明显骨化影;病理检查示肌束间组织广泛水肿,伴有轻度炎症反应,血管和成纤维细胞增生,肌束间纤维组织增生明显,并形成类骨组织和类软骨组织,局部出现较明显的异位骨化。结论　该动物模型与临床创伤性骨化性肌炎表现和国外文献一致,说明模型复制成功。Objective To set up the animal model of the traumatic myositis ossificans around the knee joints of the rabbits Methods Ten white New Zealand rabbits were strongly torn and violently massaged to result in soft tissue injuries around the knee joints Then the joints were fixed in extension position Local swelling,skin temperature,degree of ossification and local signs were observed after massaging once daily during the first week to the fourth week and alternative days from the fifth week on Pathological examination had been done in the ninth week Results It was showed that the local swelling was severe,the skin temperature was high,the wounded limb joint was stiff and the function of extension flexion was impaired obviously One can feel the ossified muscles around the stiff joint Meanwhile,ossification could be seen clearly in X ray film The extensive edema with inflammatory reaction,hyperplastic vessel,fibroblast and fibrous tissue,osteoid and chondroid tissue,obvious ossification could be seen in pathological examination Conclusion The model can mimic the clinical syndromes of traumatic myositis ossificans;the method is in agreement with that reported in domestic as well as in foreign literatures
