摘 要:利用高效液相色谱法测定柴达木枸杞叶中芦丁的含量,本文通过正交实验,对柴达木枸杞叶芦丁的提取工艺进行了优化,并对最优提取工艺条件下得到的提取物进行了纯化。结果表明:各因素对芦丁的提取效果的影响程度为乙醇浓度〉料液比〉提取温度〉超声时间。从而确定柴达木枸杞叶中芦丁的最佳提取工艺为乙醇浓度为90%,料液比为1:6(W/V),超声时温度为50℃,超声提取时间为20min,并重复提取2次。在此条件下采用AB-8大孔.树脂对提取物进行纯化..芦丁含量可达66.47%。To optivaize the extraction condition of rutin from the leaves of Lycium barbarum L. Qaidam Basin, extraction was studied by onlhogonal experiment and the concentration of rutin was determined by HPLC. And the extract was purified at the optimum condition. The results show that the order of factors to affect the rutin extraction was ethanol concen- tration 〉 solid-llquid ratio 〉 extraction temperature 〉 Ultrasonic time. The optimal means for extraction of rutin from Lycium barbarum ]eaves was to extract with 90% alcohol at 50 ℃for 20 rain, solid liquid ratio 1 : 6 for 2 times. Under these conditions, ratin content of the extract was up to 66.74% by using of AB-8 macroporous resin.
