摘  要:过氧化物酶是以过氧化氢或烷基过氧化物作为电子受体的氧化还原酶。在温和、可控条件下它可催化手性有机分子生成,再经化学酶法合成广泛的有用化合物。近年在此领域内的应用取得重要进展。位点专一性突变和定向进化是研究辣根过氧化物酶结构与功能的技术,它提供了开发工程酶的机会,这类工程酶可应用于自然产品生产、精细化学合成、医学诊断、和生物治理等。氯过氧化物酶是一种应用最广的过氧化物酶,具有’更高的手性选择性和酶稳定性。Peroxidases are ubiquitous oxidoreductases that use hydrogen peroxide or alkyl peroxides as oxidants. Advances have recently been made in using them to prepare, under mild and controlled conditions, chiral organic molecules that are valuable for the chemoenzymatic synthesis of a wide range of useful compounds. Site-directed mutagenesis and directed evolution techniques are now used routinely to investigate the structure and function of horseradish peroxidase and offer the opportunity to develop engineered enzymes for practical applications in natural product and fine chemicals synthesis ,medical diagnostics and bioremediation. A combination of horseradish peroxidase and indole-3-acetic acid or its derivatives is currently being evaluated as an agent for use in targeted cancer therapies. Chloroperoxidase, the most versatile peroxidase, behaves like a 'true' monooxygenase in sulfoxidations with molecular oxygen and an external reductant,with substantial increases in enantioselectivity and enzyme stability. Peroxidase has also been used for practical analytical applications in diagnostic kits, such as quantification of uric acid, glucose, cholesterol, lactose, etc. Enzyme- linked immunosorbent assay (ELISA) tests ,in which peroxidase enzyme is the most common enzyme used for labelling an antibody, are a simple and reliable way of detecting toxins, pathogens, cancer risk in bladder and prostate, and many other analytes.
