摘  要:通过样品处理及检测条件的优化,建立发酵虫草菌粉腺苷含量的RP-HPLC检测方法。结果表明:Agilent-C18柱(4.6 mm×300 mm,5μm),乙腈+甲醇+水=5+5+90(体积比)作为流动相,检测波长260 nm,流速为1.2 mL/min,柱温35℃,进样量为20μL,腺苷浓度在10μg/mL~50μg/mL时,浓度与峰面积呈线性关系,最低检出浓度为1.2μg/mL,定量限为4.0μg/mL,加标回收率为95.30%~105.20%,相对标准偏差为2.32%~5.70%。The methold for the determination of adenosine concentrations in the mycelia of cordyceps epiphytes by RP-HPLC was established through the optimization of sample process and detection conditions.The results showed that the chromatographic conditions included Agilent-C18 column(4.6 mm × 300 mm,5 μm) and the mobile phase consisting of acetonitrile,methanol and water by 5∶5∶90(volume ratio).The detection wavelength was at 260 nm.The flow rate was 1.2 mL/min.The column temperature was at 35 ℃.The sample dose was 20 μL.When the concentration of adenosine was at 10 μg/mL to 50 μg/mL,the perfect linearity between concentration and peak area ratio was obtained.The detection limit was 1.2 μg/mL and the quantificational limit was 4.0 μg/mL.The recovery rate was 95.30 % to 105.20 %.The relative standard deviation(RSD) was 2.32 % to 5.70 %.
