摘 要:研究了基因型、TDZ(N-苯基-N′-1,2,3-噻二唑-5-脲)和AgNO3对黄瓜离体雌核发育诱导单倍体的影响.结果表明:不同基因型黄瓜品种的雌核发育启动率不同,其变化幅度在65.52%~93.33%之间;启动雌核发育的最适TDZ浓度为0.08mg/L;诱导培养基中添加AgNO3有助于提高雌核发育启动率,其最适添加浓度为5mg/L;试验共获得8棵再生植株,流式细胞仪鉴定均为单倍体.Effect of genotype,TDZ and AgNO3 on haploid plant induced by in vitro gynogenesis was studied.The results showed that genogensis initiation frequency of different genetypes had diversity,the variation of induction frequency ranged from 65.52% to 93.33%.The optimal TDZ concentration of genogensis initiation was 0.08 mg/L in inducting medium.Adding AgNO3 to inducting medium increased genogensis initiation frequency,the optimal AgNO3 induction concentration was at 5 mg/L.Eight plants were regenerated in this research,all regenerated plants showed haploid by analysis of flow cytometry.
