摘  要:大型真菌松茸(Tricholoma matsutake)子实体经过除杂、粉碎、预处理、热水浸提、醇沉处理后,得到了松茸多糖粗提物(TMP),并运用DEAE-cellulose column和Sephadex G-100 column进行柱层析分离纯化,结果显示:以酶法与Sevage法交替法除蛋白后,可见在260nm和280nm附近均无明显吸收峰,表示除蛋白后多糖溶液中均无蛋白、核酸等杂质.从四川省小金县松茸子实体分离提取粗多糖(TMP)得率为16.4%,分离纯化后得到一种新型水溶性杂多糖(TMP-A)纯品,其得率为0.22%.The crude Tricholoma Matsutake polysaceharide(TMP) was yield after the cleaning, grinding, pre-treatment,hot water extraction, and alcohol precipitation of the fruiting bodies of Trieholoma Matsutake. The resulting polysaccharide solution was dialyzed, concentrated and lyophilized. TMP was subjected to a DEAE-cellulose column and and Sephadex G - 100 column for purity. The results showed: after the enzymatic method with Sevage method for protein clearance, it can be seen in the vicinity of 260nm and 280nm there were no significant absorption peaks, indicating that after the protein clearance the TMP has no protein, no nucleic acids and other impurities. The yield rate of TMP from Tricholorna Matsutake was 16.4% after separation and extraction processes. The yield rate of the pure product of novel water - soluble heteropolysaeeharide (TMP - A) was 0.22% after separation and purification from the starting material.
