益气活血方对实验性心肌梗死大鼠心肌梗死面积及VEGF、bFGF、Ⅷ表达量的影响

摘　　要：目的探讨益气活血方对心肌梗死大鼠冠脉血管生成作用的影响,观察其对缺血心肌的保护作用。方法应用结扎冠状动脉左前降支造成大鼠急性心肌梗死模型,观察中药参红通络浸膏对心肌梗死大鼠心脏血管新生和梗死面积的影响。探索大小剂量中药对心肌梗死后大鼠血管新生和梗死面积的作用有无不同。大鼠随机分成:参红通络浸膏高、低剂量组(A、B)、贝复济肝素对照组(C)、模型对照组(D)及假手术组(E)五组。分别对上述中药对心肌梗死大鼠心室重构的影响进行评价。结果心肌梗死4周后观察各组检测结果,参红通络浸膏高、低剂量组在梗死面积、左室梗死边缘区血管内皮生长因子(VEGF)、碱性成纤维细胞因子(bFGF)、Ⅷ因子兔源性多克隆抗体表达量以及梗死边缘区Ⅷ因子血管面密度方面,与模型组及贝复济肝素组比较,差异均有统计学意义(P<0.05);以上各指标高剂量组均优于低剂量组(P<0.05)。结论中药益气活血方具有促血管再生的作用。Objective To explore the effect of yiqi huoxue formula（the formula for benefiting qi and activating blood circulation）on coronary angiogenesis in the rats with myocardial infarction and observe the protection of the formula on ischemic myocardium.Methods The left descend branch of coronary artery was ligated to prepare the model of acute myocardial infarction in rats.The effect of shen hong tong luo extract was observed on cardiac angiogenesis and infarction area in rats.The impacts of high and low doses of Chinese medicine were explored on angiogenesis and infarction area after myocardial infarction.The rats were randomized into 5 groups,namely a high-dose Chinese medicine group（A group）,a low-dose Chinese medicine group（B group）,a control group with rb-bFGF plus heparin（C group）,a model control group（D group）and a sham-operation group（E group）.The impacts of the Chinese medicine were evaluated on the ventricular reconstruction in the rats with myocardial infarction.Results In 4 weeks of myocardial infarction,the test results were observed in each group.In terms of infarction area,the expressions of the vascular epidermal growth factor（VEGF）,the basic fibroblast growth factor（bFGF）and Ⅷ factor rabbit polyconal antibody as well as the expression of Ⅷ factor vascular surface density on the infarction margin,the results in A group and B group were significantly different in statistics as compared with D group and C group（P0.05）.All the indexes above in A group were superior to those in B group（P0.05）.Conclusion The Chinese medicine for benefiting qi and activating blood circulation promotes the angiogenesis.
