摘  要:目的:检测血管内皮功能损伤标记物纤溶酶原激活物抑制物-1(PAI-1)、血管性血友病因子(vWF)在代谢综合征幼鼠腹主动脉以及血清中的表达变化,了解有无血管内皮功能损伤。方法:3周SD幼鼠随机分为普通饮食组(NC组)、高脂饮食组(FC组)及高脂高盐饮食组(FSC组)3组,分别对体重、体长、腹围、血压、内脏脂肪重量、血脂、血糖、胰岛素等指标进行监测。采用ELISA法检测血清中PAI-1、vWF的含量,HE染色观察腹主动脉病理改变,免疫组化检测PAI-1、vWF在血管壁的表达。结果:(1)光镜检查显示NC、FC组大鼠腹主动脉内皮细胞形态正常,血管壁结构完整,而FSC组大鼠腹主动脉内皮细胞少许脱落,肌层细胞排列紊乱;(2)PAI-1、vWF在FSC组大鼠腹主动脉血管壁的表达明显高于NC、FC组(P<0.05);(3)FSC组血清中PAI-1、vWF含量明显高于NC、FC组(P<0.01)。结论:代谢综合征幼鼠已存在血管内皮功能损伤,为临床上儿童青少年代谢综合征早期关注心血管改变提供实验依据。Objective: To investigate the expression of the vascular endothelial injury markers, PAI-1 and vWF, in the abdominal aorta and the blood serum in young rats with metabolic syndrome, and evaluate the correlation of endothelial cell dysfunction with metabolic syndrome. Methods: Thirty-six 3-week-old young SD rats were randomly divided into three groups: routine diet (NC), high fat diet (FC), and high fat and salt diet (FSC). After four weeks, body weight, length, waist circumference, blood pressure, the weight of fat, the serum lipids, blood glucose and insulin were measured.In addition, the levels of PAI-1 and vWF in the serum were detected by ELISA, and the pathological changes of abdominal aorta were observed by HE staining, and the expression of PAI-1 and vWF were detected by immuno- histochemistry as well. Results: (1) No difference in the endothelial cells and the structure of the hemal wall between NC and FC group was observed. However, the endothelial cells peeled off from the abdominal aorta and muscle cells disorder were found in the FSC group compared to that of other groups. (2)The expression of PAI-1 and vWF in the abdominal aorta cells was significantly higher in the FSC group than that in other groups (P〈0.05).(3) The expression of PAI-1 and vWF in the serum was significantly higher in the FSC group than that in other groups(P〈0.05 ). Conclusion: Endothelial cell dysfunction is correlated with metabolic syndrome in the young rat model, therefore, experimental evidence is provided for early cardiovascular attention to children and adolescents with metabolic syndrome in the clinical work.
