摘  要:目的探讨锌缺乏对小鼠大脑海马锌离子和锌转运蛋白4(Zinc Transporter 4,ZnT4)表达的影响.方法建立锌缺乏动物模型,给断乳后的CD-1小鼠分别喂养缺锌饲料和正常饲料,5周后取大脑海马组织,应用锌金属自显影技术、免疫组织化学技术和Western blotting技术分别进行两组海马锌离子含量和ZnT4表达的定性定量检测.结果锌缺乏5周后,海马各区锌离子含量减少,ZnT4蛋白表达下降.结论锌缺乏能够导致海马锌离子降低和ZnT4表达下调,从而影响细胞内锌稳态,最终影响小鼠的学习记忆能力.Objective To investigate the effects of zinc deficiency on the zinc ion and zinc transporter 4 expression in hippocampal neurons.Method CD-1 mice were fed with zinc deficient diets or normal diets after weaning.The hippocampi were taken after five weeks.Autometallography,histochemistry and Western blotting were used for the detection of zinc ion content,and qualitative and quantitative detection of zinc transporter 4 expressions in the hippocampal neurons in both groups.Results The zinc ion contents were decreased in various areas of the hippocampus.And the expression of zinc transporter 4 was downregulated after zinc deficiency for 5 weeks.Conclusion The zinc deficiency might result in a decrease of zinc ion and a down-regulation expression of zinc transporter 4 in hippocampus,which may affect the homeostasis of zinc and finally affect mice's learning and memory function.
