摘  要:目的探讨雷公藤内酯醇(TP)联合紫杉醇对人卵巢癌耐顺铂细胞株(CoC1/DDP)体外活性的影响及其可能机制。方法将对数生长期CoC1/DDP细胞随机分为4组,TP组采用10 ng/ml TP作用于细胞,紫杉醇组采用3.13μg/ml紫杉醇,联合组采用10 ng/ml TP和3.13μg/ml紫杉醇,空白对照组不作任何处理(调零组),采用MTT法检测各组细胞生长抑制率,透射电子显微镜下观察细胞超微结构变化,流式细胞仪检测细胞周期及细胞凋亡情况。结果联合组生长抑制率明显高于单药组,且呈时间依赖性(P<0.05);透射电子显微镜示TP组、紫杉醇组、联合组细胞均呈凋亡改变,联合组细胞凋亡率明显高于单药组,紫杉醇组与联合组细胞周期中G2/M期的比例明显增高(P均<0.05)。结论 TP联合紫杉醇可协同抑制CoC1/DDP细胞生长,促进其凋亡,其机制可能为使细胞阻滞于G2/M期。Objective To investigate the effect of Triptolide and Paclitaxel on the activity of drug-resistant ovarian cancer cell(CoC1/DDP cells)in vitro and its possible mechanism.Methods The CoC1/DDP cells in exponential phase of growth were divided into four groups randomly,the TP group was treated by TP(10 ng/ml),the paclitaxel group was treated by paclitaxel(3.13 μg/ml),the combined group was treated by TP(10 ng/ml) and paclitaxel(3.13 μg/ml),the control group was given no treatment(zero group).MTT assay was used to detect the cell growth inhibition rate of each group,transmission electron microscope was used to observe the change in cell ultrastructure of CoC1/DDP cells,the proportion of cell cycle and the apoptic ratios of CoC1/DDP cells were measured by flow cytometry.Results The growth inhibiting rate of CoC1/ DDP cells of combined group was significantly higher than that of TP group and paclitaxel group,in time-dependent manner(P〈0.05);apoptotic cells could see in TP group,paclitaxel group and combined group under transmission electron microscope,the apoptic ratio of CoC1/DDP cells of the combined group was significantly higher than that of the single-drug group(both P〈0.05).The proportion of G2/M phase of CoC1/ DDP cells increased obviously in paclitaxel group and the combined group(P〈0.05).Conclusion There is a synergistic effect when triptolide and paclitaxel are used together to inhibit the growth and induce poptosis of CoC1/DDP cells,the mechanism may be associated with blocking cell cycle in G2/M phase.
