摘 要:目的探讨1,25-二羟基维生素D3[12,5-(OH)2D3]对大鼠肾小球系膜细胞(MC)增殖的影响及机制,为其临床应用提供依据。方法将体外培养的对数生长期大鼠MC分为对照组和观察组。观察组采用1,25-(OH)2D3(浓度为10-8mol/L)干预,对照组不干预。采用四甲基偶氮唑蓝(MTT)比色法检测细胞增殖情况,流式细胞仪检测细胞周期,免疫荧光染色法检测蛋白激酶(Akt)/哺乳动物雷帕霉素靶蛋白(mTOR)表达。结果与对照组比较,观察组MC增殖受抑,细胞周期阻滞于G1期;Akt、mTOR蛋白表达降低,P均<0.05。结论 1,25-(OH)2D3可抑制大鼠肾小球MC增殖,其作用机制可能为抑制磷脂酰肌醇-3激酶/Akt/mTOR信号传导通路。Objective To investigate the effect of 1,25-dihydroxyvitamin D3 on the proliferation of rat glomerular mesangial cells(MC) and its mechanism,in order to provide basis for the clinical application.Methods MC of rat in exponential phase were divided into observed group and control group,the observed group was treated with 1,25-(OH)2D3(10-8 mol/L),and the control group was not intervened.MTT colorimetric method was used to detect the proliferation of the cells,flow cytometry was used to test the cell cycle,immunofluorescent staining was used to determine the expression of mTOR,Akt protein.Results Compared with the control group,the proliferation of the MC was obviously inhibited in observed group,and the cell cycle block ed in the G1 phase,expression of Akt,mTOR protein reduced,all P〈0.05.Conclusions 1,25-(OH)2D3 can inhibit proliferation of MC,its mechanism may be related with the inhibition of PI3K/Akt/mTOR signal pathway.
