四川泸州翅果菊的核型研究(英文)

摘　　要：目的：研究四川泸州中药翅果菊的染色体数目和核型。方法：翅果菊种子22℃暗培养，根尖经冰水混合物预处理，卡诺氏固定液Ⅰ固定。1M HCl 60℃解离。希夫试剂染色，分生组织区用45％乙酸制片，中期细胞光镜观察照相。结果：泸州翅果菊的染色体数目为18。核型公式为2n=2x=18=6m＋10sm＋2st。染色体相对长度范围8．61-13．92，核型不对称系数为67．60。结论：四川泸州翅果菊核型类型属于3A型，不同于已报道的翅果菊居群，核型分化极可能与翅果菊物种的广布和生境不同有关。近端部着丝点表明泸州翅果菊属于较为进化的类群。Objective： To obtain the chromosome numbers and karyotype of Luzhou （Sichuan） Pterocypsela indica. Methods： Seeds of P. indica were germinated 22℃ in the dark. Root tips were pretreated in an ice bath, fixed in Carnoy＇s fixative Ⅰ , and treated in 1 M HCl at 60℃ in a water bath. Somatic cells were stained in Schiff and the meristematic portions of the root tips were squashed in 45% acetic acid. Microphotographs were taken from metaphase cells by optical microscope. Results： The chromosome numbers of P. indica were 18, and the karyotype formula was 2n= 2x= 18= 6m＋10sm＋2st. The relative length of chromosomes ranged in size from 8.61 to 13.92. The index of the karyotypic asymmetry was 67.60. Conclusion： The karyotype of Luzhou P. indica is 3A type. There are variations in chromosomal parameters and characteristics between Luzhou P. indica and other populations. The karyotype differentiations are related to their wide distributions and different habitats The subterminal centromere indicated that Luzhou P. indica is most likely more evolved populations.
