摘  要:目的观察壳聚糖温敏凝胶与富含血小板血浆(PRP)以不同比例共混后的微观结构,探讨其作为牙周组织再生支架材料的可行性。方法制备壳聚糖温敏凝胶及PRP,将二者按不同体积比共混后分为5组:A组,单纯壳聚糖温敏凝胶;B组,壳聚糖温敏凝胶:PRP=2:I;C组,壳聚糖温敏凝胶:PRP=1:1;D组,壳聚糖温敏凝胶:PRP=1:2:E组,单纯PRP。各组冷冻干燥后通过扫描电镜观察其微观结构。结果壳聚糖温敏凝胶共混不同比例的PRP后在37cc仍可形成凝胶。随着加入PRP比率增大,壳聚糖温敏凝胶的孔径逐渐增大:A组为50μm左右;B组为30~60μm;C组为100~200μm;D组为200—300μm;E组为200~500μm。结论PRP可以增大壳聚糖温敏凝胶的孔径.C组和D组的孔径已经达到比较理想的组织工程支架材料的要求。壳聚糖温敏凝与PRP共混有望作为支架材料促进牙周组织再生。Objective To investigate the micro-morphous of chitosan thermosensitive hydrogel mixed with platelet-rich plasma (PRP), and explore the feasiblity of the combination to be the scaffold material in periodontal tissue regeneration. Methods Preparing ehitosan thermosensitive hydrogel and PRP, combining them with different volume ratio and then dividing them into 5 groups (A, pure chitosan thermosensitive hydrogel; B, chitosan thermosensitive hydrogel:PRP=2:1; C, chitosan thermosensitive hydrogel:PRP=1 :1; D, chitosan thermosensitive hydrogel:PRP=1:2; E, pure PRP) were taken. Each group was observed by scanning electron microscopy after lyophilizing. Results Chitosan thermosensitive hydrogel combined with different volume ratio PRP can still gel at 37℃ With the increasing of PRP volume ratio, the aperture size of chitosan thermosensitive hydrogel grew up. The aperture size of groups(A-E) were respectively about 50μm, 30-60 μm, 100-200 μm, 200-300 μm and 200- 500μm. Conclusion PRP can increase the aperture size of chitosan thermosensitive hydrogel. The aperture sizes of group C and D have already reached the ideal demand of tissue engineering scaffold. The ehitosan thermosensitive hydrogel/PRP is likely to be used as a new scaffold material in the periodontal L regeneration.
