摘 要:[目的]对甜瓜种质资源遗传多样性进行ISSR分析。[方法]以27份甜瓜品种为材料,对其基因组DNA进行ISSR分析,计算它们之间的遗传相似系数并进行聚类分析。[结果]从69个ISSR引物中筛选出9条扩增谱带清晰、反应稳定的引物,共扩增出73条条带,其中56条为多态性条带,平均每个引物6.2条。品种间遗传相似系数在0.65~0.97间,表明这27份甜瓜品种间的遗传基础相对较狭窄。利用UPGMA聚类分析,以相似系数0.65为标准,将27份甜瓜种质划分为薄皮甜瓜和厚皮甜瓜2大类群;以相似系数0.80为标准,将薄皮甜瓜分为7个亚类群。[结论]为遗传背景相对狭窄的甜瓜品种选育工作提供了分子水平上的依据。[Objective] The aim was to analysis the genetic diversity of Cucumis melo L.based on inter simple sequence repeat(ISSR).[Method] 27 melon cultivars were used as material for the ISSR analysis,the genetic similarity coefficient(GS) between them was calculated and then a UPGMA cluster analysis was done.[Result] 9 primers were selected from 69 primers,total 73 fragments were amplified and 56 were polymorphic ones,the average of each primer was 6.2 fragments.The GS value of 27 cultivars varied from 0.65 to 0.97,indicating the genetic diversity of these cultivars was relatively low.The UPGMA cluster analysis showed the cultivars tested in this study could be divided into thin skinned melon and thick skinned melon according to GS 0.65,and divided 7 subspecies of thin skinned melon according to GS 0.80.[Conclusion] It provided the molecular basis of the melon breeding.
