氟氯西林钠胶囊在中国汉族和藏族健康人体内的药动学研究

摘　　要：目的:研究汉族和藏族健康志愿者单剂量口服氟氯西林钠胶囊的药动学。方法:健康志愿者20名(其中汉族和藏族各10名,男女各半),单剂量口服氟氯西林钠胶囊500mg,采用HPLC法测定血浆中氟氯西林的含量。用DAS 2.1软件进行数据处理,用SPSS软件对不同性别志愿者的药动学参数进行统计分析。结果:单剂量口服500mg氟氯西林钠胶囊后,汉族和藏族志愿者的主要药动学参数如下:cmax为(16.85±7.13)和(14.23±6.04)μg/ml,tmax为(1.02±0.07)和(0.95±0.04)h,t1/2为(1.61±0.24)和(1.45±0.29)h,AUC0～10为(61.36±15.63)和(63.02±17.74)μg.h.ml-1,AUC0～∞为(65.74±16.87)和(64.36±19.54)μg.h.ml-1。男性和女性志愿者的主要药动学参数如下:cmax为(16.93±6.16)和(14.18±7.83)μg/ml,tmax为(0.93±0.04)和(1.05±0.06)h,t1/2为(1.58±0.35)和(1.44±0.21)h,AUC0～10为(61.85±17.62)和(63.64±14.75)μg.h.ml-1,AUC0～∞为(63.52±16.73)和(65.36±20.04)μg.h.ml-1。结论:健康志愿者单剂量口服氟氯西林钠胶囊的体内药动学过程符合二室开放模型。经统计学分析,氟氯西林在汉族和藏族健康志愿者体内的药动学过程无显著性种族差异和性别差异。Objective： To study the pharmacokinetics of flucloxacillin sodium capsules after a single oral dose in Chinese Han and Tibetan healthy volunteers. Methods：Ten Han and ten Tibetan healthy volunteers were involved in the study. They were evenly composed of male and female. Each subject received a single oral dose of 500 mg flucloxacillin sodium capsules. The plasma concentration of flucloxacillin was determined by HPI.C method. The pharmacokinetic parameters were calculated by DAS 2.1 software and were statistically analyzed by SPSS software. Results：The main pharmacokinetic parameters of fluclox aeillin in Han and Tibetan volunteers were as follows： cmax,（16.85±7. 13） and （14.23±6.04） μg/ml,tmax （1.02±0.07） and （0.95±0.04） h,t1/2（l.61±0.24） and （1.45±0.29） h,AUC010（61.36±15. 63） and （63.02±17.74） μg. h. ml^-1,AU（L （65.74±16.87） and （64.36±19.54） μg · h· ml^-1.These parameters of male and female volunteers were as follows： cmax （16.93±6.16） and （14.18±7.83） μg/ml,tmax（0. 93±0.04） and （1.05±0.06） h,t1/2 （1.58±0.35） and （1.44±0.21） h, AUC0-10 （61.85±17.62） and （63.64±14.75） μg. h. ml ^-1,AUC0-10 （63.52±16.73） and （65.36±20.04） μg h. ml L Conclusion：The plasma concentration-time curve of flucloxacillin meets a two compartment open model,and its main pharmaco kinetic parameters have no significant differences between Han and Tibetan nationality,or male and female volunteers.
