摘  要:目的活体监测Gd-DTPA标记的脂肪干细胞(ADSC)在退变椎间盘内的存活、迁移和转归,观察ADSC对修复及延缓退变的作用。方法将新西兰大白兔30只制成腰椎椎间盘退变模型,2周后行Gd标记的ADSC(Gd-ADSC)移植及ADSC移植,在不同时间点摄X线片、行MR扫描及病理学检查,并与移植PBS比较。结果 Gd-ADSC移植后即刻,SPIR T1WI显示椎间盘内高信号随时间推移迅速减低。与同时间点移植PBS比较,ADSC及Gd-ADSC移植兔椎间盘退变程度较轻,髓核及纤维环内均可见胞浆丰富软骨样细胞。结论 ADSC椎间盘内移植有助于修复退变椎间盘和延缓椎间盘退变;Gd-ADSC移植后1周内可通过MRI进行监测。Objective To investigate the contribution of ADSC to repair intervertebral degenerative disc (IDD), and to observe the in vivo tracking of transplanted Gd-DTPA labeled ADSC survive and migration by MRI. Methods Thirty Newland white rabbits underwent transplantation of Gd-DTPA labeled ADSC 2 weeks after of the establishment of IDD mod- els. X-ray films, MRI and histopathological results were obtained in time interval after transplantation of Gd-DTPA labeled ADSC and ADSC, and were compared with those of transplantation of PBS. Results High signal intensity on SPIR T1WI was detected right after intervertebral disc transplantation of Gd-DTPA labeled ADSC, but the signal intensity decreased rapidly with time going. Degeneration was mild after ADSC and Gd-DTPA labeled ADSC transplantation, and Chondrocyte-like cells was apparent in the intervertebral disc. Conclusion ADSC transplantation is effective in regeneration degenerated intervertebral disc. Gd-DTPA labeled ADSC can be identified by MRI within 1 week.
