摘  要:目的探讨肺动脉粥样硬化模型兔肺动脉壁骨桥蛋白(OPN)表达的变化。方法 24只新西兰兔,随机平均分为正常对照组和高脂模型组,分别给予基础饲料和高脂饲料(基础饲料+1%胆固醇+5%猪油)喂养12周后取血和肺动脉,分析血清中相关脂质成分的变化;HE染色观察肺动脉壁的形态结构;免疫组化法和Western blot检测肺动脉粥样硬化兔肺动脉壁OPN的表达。结果成功建立肺动脉粥样硬化兔模型,高脂模型组在高脂饮食12周后,与正常对照组比较,血清中甘油三酯(TG)、胆固醇(CHO)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)有明显增加(P<0.05),其中以LDL和LDL/HDL比值升高最明显;HE染色显示高脂模型组肺动脉内膜增厚,细胞间隙逐渐增大,泡沫细胞形成,可见明显的动脉粥样硬化斑块;免疫组化法和Western blot显示高脂模型组肺动脉壁OPN的表达较正常对照组显著增强,差异有统计学意义(P<0.05)。结论高脂饮食可导致肺动脉粥样硬化形成,OPN在肺动脉粥样硬化斑块形成中起重要的作用。Objective To investigate the osteopontin (OPN) expression of pulmonary artery in pulmonary artery atherosclerosis model rabbit. Methods 24 New-Zealand rabbits were randomly divided into normal control group and high-fat diet model group. They fed with normal diet and high-fat diet respectively for 12 weeks. The serum level of the lipids was analyzed. The OPN expression of the pulmonary artery wall was detected by immunohistochemistry and Western blot. The morphology of pulmonary artery wall was examined by HE stain. Results The pulmonary artery atherosclerosis rabbit model was established successfully. Compared with the normal control group,the serum levels of CHO, LDL, HDL, TG, LDL/HDL in high-fat diet group were obviously increased( P 〈 0.05 ). Immunohistochemistry and Western blot showed that the OPN protein overexpressed in pulmonary artery wall in model rabbits(P 〈0.05). Meanwhile, the thickening of tunica intima, intercellular space increased gradually, formation of foam cells, and atheroselerosis plaque could be observed in model rabbits by HE staining. Conclusion High fat diet can induce the pulmonary artery atherosclerosis. OPN may play an important role in the development of pulmonary artery atheroselerosis.
