摘  要:目的观察负压封闭引流技术(VSD)对创面细胞黏附分子-1(ICAM-1)表达及IκBα磷酸化的影响。方法每组建立24例Susscrafa猪背急性感染创面模型,VSD治疗为实验组,120mmHg(1mmHg=0.133kPa)负压持续吸引,常规换药治疗为对照组,对创面炎性反应、ICAM—1表达及IκBα磷酸化的水平进行分析。结果与对照组比较,VSD组减轻感染创面炎性反应,ICAM-1表达相对值0.60±0.08,显著低于对照组1.01±0.06(P〈0.05),VSD显著减少IκBα磷酸化(40±9)%,(P〈0.05)。结论负压封闭引流减少IκBα磷酸化,抑制感染创面ICAM-1的表达,明显减轻感染创面炎性反应。Objective To investigate the effect of vacuum sealing drainage (VSD) on intercellular adhesion molecule-1 ( ICAM-1 ) and related mechanism in wound healing. Methods Models of acute infected wound were made in each 24 standard samples on back of Sus scrafa pigs, and VSD was performed as treated group with -120 mm Hg (1 mm Hg = 0. 133 kPa) , while common dressing as control group. Wound was assessed by Hematoxyiin and Eosin (HE) stain at the 7th day after treatment. The expression of ICAM-1 was detected by using immunochemistry, real-time polymerase chain reaction (PCR) and Western blotting in granulation tissues as well as the p-IκBα and IκBα by using Western blotting. Results ICAM-1 in the VSD group was 0. 60±0. 08, lower than the control group ( 1.01 ±0. 06) , P 〈 0. 05 ; the protein level of p-IκBα was decreased by (40 +9)% as well as p-IκBα/IκBα in VSD group as compared with control group (P 〈 0. 05). Conclusion VSD obviously decreases the expression of ICAM-1 via suppressing IκBα phosphorylation in the wound healing.
