摘 要:目的:观察溃疡性结肠炎(UC)患者血清SOCS-3、IL-6、TNF-α水平变化,探讨其在UC发生发展中的关系及意义.方法:选取UC患者(n=59),其中活动期(n=46),缓解期(n=13),活动期患者中,轻度(n=15),中度(n=24),重度(n=7).采用ELISA法检测各组血清IL-6、TNF-α、SOCS-3含量,并和20例正常体检者进行对照.结果:UC活动组患者血清中IL-6、TNF-α含量均明显高于对照组及缓解组(P<0.01),而血清SOCS-3含量显著低于对照组及缓解组(P<0.01);活动组中,轻、中、重度患者血清IL-6、TNF-α含量随严重程度逐渐升高,血清SOCS-3含量逐渐降低,三级之间差异均具有统计学意义(P<0.05,P<0.01);相关性分析表明,血清IL-6含量与TNF-α含量呈明显正相关(r=0.817,P<0.01);血清SOCS-3含量与IL-6、TNF-α含量均呈明显负相关(r=-0.667、-0.611,P<0.01).结论:UC患者血清SOCS-3水平明显降低,其可能通过负性调节IL-6、TNF-α参与UC的发生发展,且与病情的严重程度有关.AIM:To investigate the changes in the levels of serum suppressor of cytokine signaling-3 (SOCS-3),interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) in patients with ulcerative colitis(UC) and to analyze the role of SOCS-3, IL-6 and TNF-αin the genesis and development of ulcerative colitis(UC). METHODS:According to the severity of UC, 59 patients with UC were divided into active group(n = 46) and remission group(n = 13). The numbers of patients with mild,moderate, and severe UC in the active group were 15,24 and 7,respectively.Twenty healthy individuals served as normal controls.The levels of serum SOCS-3,IL-6 and TNF-αwere determined by ELISA. RESULTS:The levels of serum IL-6 and TNF-αwere significantly higher and that of serum SOCS-3 was significantly lower in patients in the active group than in those in the remission group and control group(all P 0.01).The levels of serum IL-6 and TNF-αgradually increased and that of serum SOCS-3 gradually decreased with the aggravation of UC.Significant differences were found in serum levels of SOCS-3, IL-6 and TNF-αamong patients with mild, moderate or severe disease(all P 0.05).A positive correlation was noted between the levels of serum IL-6 and TNF-α(r = 0.817,P 0.01),and a negative correlation was observed between serum level of SOCS-3 and those of IL-6 and TNF-α. CONCLUSION:A decrease in serum levels of SOCS-3 in patients with UC may correlate with the progression of UC by negatively regulating serum levels of IL-6 and TNF-α.
