摘  要:目的探讨丹参酮ⅡA对低氧培养的人高转移性巨细胞肺癌(PG)细胞表面黏附分子E-cadherin、β1-整合素表达的影响。方法 PG细胞复苏,按干预药物不同分为丹参酮ⅡA高、中、低剂量组及顺铂组、阴性对照组,分别置于常氧和低氧环境下培养,48 h后应用流式细胞术测定黏附分子E-cadherin、β1-整合素在各组PG细胞表面表达情况。结果低氧环境下,PG细胞所表达β1-整合素荧光强度随丹参酮ⅡA浓度的增高而逐渐减弱,但表达β1-整合素的阳性细胞率接近100%,无统计学意义;PG细胞表达E-cadherin,随着丹参酮ⅡA浓度的增加,其阳性细胞数增加,但表达E-cadherin的荧光强度很弱,无统计学意义。常氧环境下,PG细胞所表达β1-整合素的荧光强度随着丹参酮ⅡA浓度的增高逐渐减弱,表达β1-整合素的阳性细胞率接近100%,无统计学意义;PG细胞表达E-cadherin,随着丹参酮ⅡA浓度的增加,其阳性细胞数减少,但表达E-cadherin荧光强度很弱,没有统计学意义。结论在常氧环境下,丹参酮ⅡA通过下调E-cadherin和β1-整合素表达来抑制PG细胞的浸润转移;在低氧环境下,通过下调β1-整合素来抑制PG细胞的转移而上调E-cadherin表达来增加PG细胞的浸润转移能力。Objective To investigate the effect of Tanshinon ⅡA on the expression of β1-integrin and E-cadherin on the surface of cell PG cultured during normoxia and hypoxia. Methods The revival cell PG was divided into three dosages (high, medium and low) group of Tanshinon ⅡA, DDP group and negative control group. All of five groups were cultured separately in the normal and hypoxic environment. The expression of E-cadherin and β1-integrin on the surface of PG cell by the flow cytometry 48 h later. Results In the hypoxic environment, the fluorescence intensity of β1-integrin increased with the density of Tanshinon ⅡA but the expression of fluorescence intensity was fading gradually. However, the rate of positive cells of β1-integrin was almost 100%, with no statistical significance. The expression of E-cadherin was increased with the density of Tanshinon ⅡA. The amount of positive cell was rising, but the fluorescence intensity of E-cadherin was very weak, with no statistical significance. In the normal oxygen environment, the fluorescence intensity of β1-integrin increased with the density of Tanshinon ⅡA, but the rate of positive cells of β1-integrin was almost 100%, with no statistical significance. The expression of E-cadherin was increased with the density of Tanshinon ⅡA. The amount of positive cell was decreasing, but the fluorescence intensity of E-cadherin was very weak, with no statistical significance. Conclusion In the normoxia, Tanshinon ⅡA restrains the expression of E-cadherin and β1-integrin to inhibit the matastasis. In the hypoxic environment, Tanshinon ⅡA restrains the β1-integrin to inhibit the matastasis and promote the expression of E-cadherin to strengthen the ability of invasion and matastasis of cell PG.
