摘 要:目的观察地黄益智方对Aβ1-40所致老年性痴呆模型大鼠细胞周期蛋白异常表达的影响,并探讨其机制。方法 120只SD雄性大鼠随机分为空白组、模型组及地黄益智方小、中、大剂量组。采用大鼠双侧海马CA1区注射Aβ1-40复制老年性痴呆模型,造模24 h后开始灌胃给药,地黄益智方小、中、大剂量组给药量分别为0.12、0.24、0.48 g浸膏/(100 g.d),空白组和模型组给予等体积的蒸馏水,连续4周。免疫组化法动态检测各组大鼠大脑皮层及海马cyclin D1、cyclin E、p16、CDK6的表达变化。结果 Aβ1-40造模后24 h可见cyclin D1、CDK6表达升高,造模后2周时cyclin E表达升高,造模后4周时p16表达升高;小、中、大剂量地黄益智方可改善Aβ1-40诱导cyclin D1、cyclin E、p16以及CDK6异常表达。结论老年性痴呆模型大鼠早期存在脑细胞周期蛋白异常表达现象,地黄益智方对异常表达的cyclin D1、cyclin E、p16及CDK6有一定改善作用。Objective To observe the effect of Dihuang Yizhi Fang on cell cycle related gene expression in senile dementia rats induced by Aβ1-40. Methods One hundred and twenty Sprague-Dawley rats were randomly divided into normal, model, low-dose, middle dose and high-dose of Dihuang Yizhi Fang group. Senile dementia was induced by Aβ1-40 injection in bilateral hippocampal CA1 area of these rats. After 24 h of Aβ1-40 injection, rats were treated intragastrically with Dihuang Yizhi Fang at concentration of 0.12, 0.24, 0.48 g extract/(100 g·d) in low-dose, middle dose and high-dose group respectively, and equal volume of distilled water in normal and model group for 4 weeks. Expression of cyclin D1, cyclin E, p16 and CDK6 in cortex and CA1 hippocampus zone were detected by immunohistochemistry. Results Expression of cyclin D1 and CDK6 were up-regulated after 24 h of Aβ1-40 injection, cyclin E expression up-regulated in 2nd weeks, and p16 expression up-regulated in 4th weeks. Low-dose, middle dose and high-dose of Dihuang Yizhi Fang may ameliorate the abnormal expression of cyclin D1, cyclin E, p16 and CDK6 induced by Aβ1-40. Conclusion Cell cycle related genes abnormally expressed in early senile dementia rats, and Dihuang Yizhi Fang may ameliorate the abnormal expression of cyclin D1, cyclin E, p16 and CDK6.
