摘  要:目的检测NGF及其受体TrkA和p75在发情周期内Wistar大鼠卵巢内的表达。方法利用免疫组织化学染色技术检测NGF及其受体TrkA和p75在卵巢内的细胞定位及蛋白表达情况;利用原位杂交方法检测在发情周期内大鼠卵巢NGF及其受体TrkA和p75的mRNA表达情况;利用Western blot技术检测发情周期内大鼠卵巢NGF及其受体TrkA和p75的蛋白表达情况。结果 免疫组织化学染色结果显示发情周期内大鼠卵巢内NGF及其受体的表达,在各级卵泡的卵泡细胞、卵母细胞以及黄体内均有不同水平的表达;同时间点的原位杂交检测结果与免疫组织化学染色结果相一致;Western blot检测结果显示整个发情周期内卵巢内的NGF及其受体TrkA和p75的蛋白表达没有明显的波动。结论NGF及其受体TrkA和p75可促进发情周期内Wistar大鼠卵巢卵泡的卵泡细胞及卵母细胞的发育。Objective To detect the expression of NGF and its receptors TrkA and p75 in Wistar rats ovary during the estrous cycle. Methods Immunohistochemistry for the localization and protein expression of NGF and its receptors TrkA and p75 in ovarian cells; In situ hybridization for the mRNA expression of NGF and its receptors TrkA and p75 in the ovary during the estrous;Western blotting for the protein amount of NGF and its receptor TrkA and p75 in the ovary during the estrous. Results The results of immunohisochemistry showed there were different expression levels of NGF and its receptor TrkA and p75 in the ovary,included the follicles, oocytes and corpus luteum in the ovary during the estrous ; the results of in situ hybridization during the same period showed the same mRNA levels change of them as immunobistochemsitry results showed;but the results of Western blot during the same period showed there were not obviously change of the protein amount change of NGF and its receptor TrkA and p75. Conclusion NGF and its receptors TrkA and p75 can promote the development of follicle cells and oocytes in the Wistar rat's ovary during the estrous cycle.
